芳香精油通過嗅覺通路調(diào)節(jié)大鼠血壓的實(shí)驗(yàn)研究

【摘要】: 目的 通過大鼠吸入3種精油來觀察其對血壓的影響,并結(jié)合嗅上皮毀損模型,以及免疫組織化學(xué)方法檢測不同腦區(qū)的神經(jīng)遞質(zhì)變化,探討精油調(diào)節(jié)血壓的可能機(jī)制,尋找新的降低血壓的藥物和作用途徑,為臨床治療高血壓病提供實(shí)驗(yàn)依據(jù)。 方法 本研究首先將大鼠隨機(jī)分為空白組、薰衣草精油組、迷迭香精油組和丁香精油組,采用MedLab 6.0生物信號采集處理系統(tǒng)測定大鼠血壓,通過實(shí)驗(yàn)篩選出具有降壓作用的薰衣草精油。再取大鼠,隨機(jī)分為空白組、薰衣草精油組、假手術(shù)+薰衣草精油組和模型(嗅上皮毀損)+薰衣草精油組,其中嗅上皮毀損模型采用硫酸鋅(ZnSO4)制備。利用MedLab 6.0生物信號采集處理系統(tǒng)測定大鼠血壓,并采用酶聯(lián)免疫吸附法測定外周血兒茶酚胺(CA)含量;通過免疫組織化學(xué)方法檢測各組大鼠腦干、下丘腦、杏仁核等腦區(qū)的γ-氨基丁酸(GABA)、5-羥色胺(5-HT)、去甲腎上腺素(NE)神經(jīng)遞質(zhì)變化。 結(jié)果 1.薰衣草精油組、迷迭香精油組、丁香精油組和空白組4組間SP、DP、MAP的差異均有顯著性?瞻捉M在吸入蒸餾水結(jié)束后100 min內(nèi)SP、DP、MAP無明顯變化,差異無顯著性。與空白組比較,薰衣草精油組SP、DP、MAP降低,在薰衣草精油吸入結(jié)束后80~100 min時降低最明顯,差異有顯著性,在觀察期100 min內(nèi)血壓未回復(fù)到干預(yù)前水平。而迷迭香精油組、丁香精油組SP、DP、MAP與空白組比較,差異無顯著性。與薰衣草精油組比較,迷迭香精油組、丁香精油組SP、DP、MAP無明顯降低,其差異具有顯著性。迷迭香精油組和丁香精油組SP、DP、MAP相比差異無統(tǒng)計(jì)學(xué)意義。 2.空白組、薰衣草精油組、假手術(shù)+薰衣草精油組和模型(嗅上皮毀損)+薰衣草精油組4組比較,4組間SP、DP、MAP差異有顯著性。與空白組比較,薰衣草精油組、假手術(shù)+薰衣草精油組SP、DP、MAP均降低,在吸入精油結(jié)束后80~100 min差異有顯著性;而模型+薰衣草精油組SP、DP、MAP無明顯降低,差異無顯著性。與模型+薰衣草精油組相比,假手術(shù)+薰衣草精油組SP、DP、MAP降低,在吸入精油結(jié)束后80~100 min差異有顯著性。薰衣草精油組、假手術(shù)+薰衣草精油組SP、DP、MAP相比差異無統(tǒng)計(jì)學(xué)意義。 3.與吸入前比較,薰衣草精油組在吸入結(jié)束后100 min血清CA濃度明顯降低,其差異具有統(tǒng)計(jì)學(xué)意義;而空白組、模型+薰衣草精油組在吸入前后血清CA濃度均無明顯變化,差異無顯著性。吸入前,各組血清CA濃度差異無統(tǒng)計(jì)學(xué)意義。吸入后,與空白組比較,薰衣草精油組血清CA濃度降低,差異有顯著性;而模型+薰衣草精油組血清CA濃度差異無統(tǒng)計(jì)學(xué)意義。 4.免疫組織化學(xué)結(jié)果顯示,與空白組比較,薰衣草精油組大鼠的杏仁核、下丘腦、腦干核團(tuán)內(nèi)5-HT、GABA表達(dá)增加,而NE表達(dá)減少。模型+薰衣草精油組與空白組比較,5-HT、GABA、NE的表達(dá)無明顯差異。 結(jié)論 1.大鼠吸入薰衣草精油能降低SP、DP、MAP; 2.薰衣草精油通過嗅覺途徑降低大鼠SP、DP、MAP; 3.大鼠吸入薰衣草精油降低外周血兒茶酚胺含量可能是其降低血壓的外周作用機(jī)制; 4.薰衣草精油通過嗅覺通路引起大鼠腦內(nèi)GABA、5-HT、NE神經(jīng)遞質(zhì)的改變可能是其降低血壓的中樞作用機(jī)制。
[Abstract]:objective
The effects of 3 kinds of essential oils on blood pressure were observed in rats, and the changes of neurotransmitters in different brain regions were detected in combination with the damage model of olfactory epithelium and immunohistochemical method. The possible mechanism of essential oil regulating blood pressure was explored, and a new drug and path to reduce blood pressure were found, and the experimental basis for clinical treatment of hypertension was provided.
Method
In this study, rats were randomly divided into blank group, lavender essential oil group, rosemary essential oil group and lilac essential oil group. The MedLab 6 biological signal collection and treatment system was used to determine the blood pressure of rats. The experiment was used to select lavender essential oil with antihypertensive effect. Then the rats were randomly divided into blank group, lavender essential oil group, sham operation + lavender. Essential oil group and model (olfactory epithelium damage) + lavender essential oil group, of which the olfactory epithelium damage model was prepared by zinc sulfate (ZnSO4). The blood pressure of rats was measured by MedLab 6 biological signal acquisition and treatment system, and the content of catecholamine (CA) in peripheral blood was measured by enzyme linked immunosorbent assay, and the brainstem of each group was detected by immunohistochemistry. The changes of GABA, 5-, 5-HT and NE in the hypothalamus, amygdala and other brain regions.
Result
1. the SP, DP, MAP of lavender essential oil group, rosemary essential oil group, lilac essential oil group and blank group have significant difference. There is no significant change in SP, DP, MAP in 100 min after the end of distilled water. Compared with the blank group, lavender essential oil group SP, DP, MAP decrease, and 80~100 min after lavender essential oil is inhaled at the end of the lavender essential oil. The difference was significant. The blood pressure did not return to the pre intervention level in the 100 min observation period. There was no significant difference in the difference between the rosemary oil group and the SP, DP and MAP in the lilac essential oil group and the lavender essential oil group, and the Rosmarinus essential oil group, the SP, DP, and MAP in the lilac essential oil group were not significantly reduced, and the difference was significant. Rosemary essence was significant. There was no significant difference in SP, DP and MAP between oil group and clove essential oil group.
2. blank group, lavender essential oil group, sham operation + lavender essential oil group and model (olfactory epithelium damage) + lavender essential oil group 4 groups, the difference between the 4 groups SP, DP, MAP was significant. Compared with the blank group, lavender essential oil group, sham operation + lavender essential oil group SP, DP, MAP decreased, and the difference of 80~100 min after the end of the essential oil was significant; and model + The SP, DP, MAP of lavender essential oil group did not decrease significantly, and the difference was not significant. Compared with the model + lavender essential oil group, the sham operation + lavender essential oil group SP, DP, MAP decreased, and the difference in 80~100 min after the end of the essential oil was significant. Lavender essential oil group, sham operation + lavender essential oil group had no statistical significance compared with SP, DP, MAP.
3. compared with before inhalation, the concentration of serum CA in the lavender essential oil group decreased significantly at the end of the inhalation of 100 min, and the difference was statistically significant. But in the blank group, there was no significant change in serum CA concentration in the model + lavender essential oil group before and after inhalation. There was no significant difference in the concentration of serum CA before inhalation. After inhalation, the concentration of serum was not statistically significant. Compared with white group, the concentration of serum CA in lavender essential oil decreased, but there was no significant difference in serum CA concentration in model + lavender essential oil group.
4. immunohistochemical results showed that, compared with the blank group, the expression of 5-HT, GABA in the amygdala, hypothalamus and brainstem nuclei increased in the amygdala, hypothalamus and brainstem nuclei of lavender group. The expression of 5-HT, GABA and NE was not significantly different from that of the model + lavender essential oil group and the blank group.
conclusion
The 1. rats inhaled lavender essential oil could reduce SP, DP and MAP.
2. lavender essential oil reduces SP, DP and MAP in rats by olfactory pathway.
3. the decrease of catecholamine in peripheral blood by inhaling lavender essential oil may be the peripheral mechanism of lowering blood pressure.
4. the change of GABA, 5-HT and NE neurotransmitters in rat brain induced by olfactory pathway may be the central mechanism of lavender essential oil to reduce blood pressure.
