【摘要】：目的探索更年期高血壓燥證大鼠模型的建立方法。方法 2014年1—3月選取雌性自發(fā)性高血壓大鼠(SHR)12只,采用摸球法將SHR隨機(jī)分為更年期高血壓組和更年期高血壓燥證造模組,每組6只。采用外科手術(shù)法摘除兩組大鼠雙側(cè)卵巢,制備更年期高血壓大鼠。對(duì)更年期高血壓燥證造模組大鼠施加人工燥證環(huán)境,飼喂西北燥證食物進(jìn)行造模。觀察大鼠生物學(xué)特征(毛色、精神狀態(tài)、興奮度、進(jìn)食量、飲水量、體質(zhì)量、大小便性狀、舌象、爪象等)、血清神經(jīng)遞質(zhì)〔促腎上腺皮質(zhì)激素釋放激素(CRH)、促腎上腺皮質(zhì)激素(ACTH)、去甲腎上腺素(NE)、多巴胺(DA)、5-羥色胺(5-HT)〕及激素〔雌二醇(E2)、睪酮(T)〕水平和臟器(肝臟、脾臟、腎上腺、下丘腦)組織結(jié)構(gòu)。結(jié)果更年期高血壓燥證造模組大鼠較更年期高血壓組體質(zhì)量降低,飲水量增加,尿量減少(P0.05)。更年期高血壓燥證造模組大鼠血清CRH、NE、DA、5-HT水平較更年期高血壓組升高(P0.05)。兩組大鼠ACTH水平比較,差異無統(tǒng)計(jì)學(xué)意義(P0.05)。更年期高血壓燥證造模組大鼠較更年期高血壓組血清E2、T水平降低,E2/T升高(P0.05)。兩組大鼠肝臟、脾臟、腎上腺、下丘腦組織結(jié)構(gòu)均發(fā)生明顯改變。結(jié)論通過對(duì)更年期高血壓大鼠施加人工燥證環(huán)境,給予西北燥證食物,控制飲水量及進(jìn)食量,可成功建立更年期高血壓燥證大鼠模型。
[Abstract]:Objective to explore the method of establishing the rat model of menopausal hypertension dryness syndrome. Methods from January to January 2014, 12 female spontaneously hypertensive rats were selected. SHR was randomly divided into menopausal hypertension group and menopausal hypertension dryness model group with 6 rats in each group. The menopausal hypertensive rats were prepared by surgical excision of bilateral ovaries. The rats of climacteric hypertensive dryness syndrome model group were fed with the food of northwest dryness syndrome. The biological characteristics (color, mental state, excitability, food intake, drinking water, body mass, fecal and urinal characteristics, tongue picture) of rats were observed. The levels of serum neurotransmitters (CRH, ACTH, NE, 5-HT) and hormone (estradiol, estradiol, testosterone) and visceral organs (liver, spleen, adrenal gland, adrenocorticotropic hormone (ACTH), noradrenaline (NE), dopamine (DA), 5-hydroxytryptamine (5-HT)) and the organs (liver, spleen, adrenal gland), and the levels of serum neurotransmitters (estradiol, estradiol, estradiol, testosterone) and organs (liver, spleen, adrenal gland), Hypothalamus). Results compared with the climacteric hypertension group, the body weight, drinking water and urine volume of the climacteric hypertensive dryness model group were lower, and the urine volume decreased than that of the climacteric hypertension group (P 0.05). Compared with climacteric hypertension group, serum CRHNE-DA-5-HT level in model group was higher than that in climacteric hypertension group (P 0.05). There was no significant difference in ACTH level between the two groups (P 0.05). Compared with the climacteric hypertension group, the serum E _ 2 / T level in the climacteric hypertensive dryness model group was lower than that in the climacteric hypertension group. The structure of liver, spleen, adrenal gland and hypothalamus in both groups were obviously changed. Conclusion the rat model of menopausal hypertension dryness syndrome can be successfully established by applying artificial dryness syndrome environment to climacteric hypertensive rats, giving food to northwest dryness syndrome, controlling the quantity of drinking water and eating quantity.
