【摘要】：目的研究Axl和Mer受體是否參與紅細(xì)胞分化的調(diào)控。方法單基因(AXL-/-和MER-/-)和雙基因(AXL-/-MER-/-)敲除小鼠,利用組織切片觀察小鼠骨髓、肝臟和脾臟的組織結(jié)構(gòu);計(jì)數(shù)小鼠單根股骨內(nèi)骨髓細(xì)胞的數(shù)量;用實(shí)時(shí)定量PCR檢測(cè)與紅細(xì)胞分化相關(guān)基因mRNA的表達(dá)。結(jié)果雙基因敲除的小鼠單根股骨骨髓內(nèi)總細(xì)胞、單個(gè)核細(xì)胞和成熟紅細(xì)胞的數(shù)量分別下降了40%、34%和53%(P0.01);骨髓細(xì)胞排列稀疏,血竇內(nèi)、外成熟紅細(xì)胞數(shù)量明顯減少;雖然外周血中紅細(xì)胞數(shù)量和血紅蛋白的含量沒(méi)有改變,但存在著明顯的髓外造血,肝臟里出現(xiàn)了造血島,脾臟中紅髓明顯地增生。而單基因敲除小鼠骨髓中紅細(xì)胞數(shù)量以及組織結(jié)構(gòu)未見(jiàn)改變。且雙基因敲除小鼠中紅系細(xì)胞內(nèi)GATA-1和EpoR的mRNA表達(dá)分別降低了65%和75%(P0.01)。結(jié)論 Axl和Mer受體通過(guò)GATA-1和EpoR來(lái)調(diào)節(jié)紅細(xì)胞的分化,并且功能上存在重疊、過(guò)剩和代償。
[Abstract]:Objective to investigate whether Axl and Mer receptors are involved in the regulation of erythrocyte differentiation. Methods single gene (AXL-/- and MER-/-) and double gene (AXL-/-MER-/-) knockout mice were used to observe the tissue structure of bone marrow, liver and spleen of mice by tissue section. The number of bone marrow cells in the single femur of mice was counted, and the differentiation phase of red blood cells was detected by real-time quantitative PCR. The results showed that the number of total cells in bone marrow, mononuclear cells and mature red cells in single root bone marrow of mice with double gene knockout decreased by 40%, 34% and 53% (P0.01), and the number of bone marrow cells was sparse, and the number of mature red cells in the blood sinuses decreased significantly, although the number of red blood cells and the content of hemoglobin in the peripheral blood were not. However, there were obvious extramedullary hematopoiesis, the hematopoietic island in the liver, the red pulp in the spleen were obviously proliferated, and the number of red cells and the structure of the bone marrow in the single gene knockout mice were not changed. The mRNA expression of GATA-1 and EpoR in the erythroid cells in the double gene knockout mice was reduced by 65% and 75% (P0.01). Conclusion Axl and Mer were found. The body regulates the differentiation of red blood cells through GATA-1 and EpoR, and there are overlaps, excess and compensatory functions.
