【摘要】：有絲分裂后期促進(jìn)復(fù)合物APC/C是一個(gè)多亞基泛素連接酶E3，它在細(xì)胞周期的調(diào)控過(guò)程中起著重要的作用，APC/C主要通過(guò)兩個(gè)含有WD40重復(fù)序列的接頭蛋白Cdc20和Cdh1與底物進(jìn)行結(jié)合。Cdc20通過(guò)泛素化降解重要的細(xì)胞周期調(diào)控因子參與細(xì)胞周期的調(diào)控，Cdc20主要通過(guò)泛素化降解細(xì)胞周期蛋白B1和Seruin使姐妹染色單體分離，起始著細(xì)胞周期由中期向后期的轉(zhuǎn)化。然而，，Cdh1在細(xì)胞周期的早期是沒(méi)有活性的，APCCdh1主要在有絲分裂的末期和G1期起重要作用，它在G1期的作用被認(rèn)為是阻止未成熟的細(xì)胞過(guò)早的進(jìn)入S期。 前期研究結(jié)果表明Cdh1不僅在G1/S期的轉(zhuǎn)化過(guò)程中起著重要作用，它同時(shí)在與細(xì)胞周期無(wú)關(guān)的生命活動(dòng)中起著重要作用。由于Cdh1的缺失具有胚胎致死性，這限制了Cdh1的體內(nèi)功能研究；為了研究Cdh1在胚胎形成后期以及后天發(fā)育過(guò)程中的作用，本課題采用了Cdh1條件性敲除小鼠作為研究對(duì)象。本課題將通過(guò)Cdh1條件性敲除小鼠來(lái)研究Cdh1在小鼠肝臟再生中的作用。 本課題研究發(fā)現(xiàn)Cdh1在小鼠肝再生過(guò)程中起著重要作用，Cdh1的缺失將促使肝實(shí)質(zhì)細(xì)胞較早的通過(guò)G1/S期檢測(cè)位點(diǎn)，使肝實(shí)質(zhì)細(xì)胞過(guò)早的進(jìn)入S期，但在肝再生過(guò)程中，Cdh1的缺失對(duì)小鼠的肝重體重比沒(méi)有顯著影響。進(jìn)一步研究表明，Cdh1的缺失對(duì)小鼠肝實(shí)質(zhì)細(xì)胞的形態(tài)并無(wú)影響，此外研究還表明盡管Cdh1的缺失能夠促使肝實(shí)質(zhì)細(xì)胞過(guò)早的通過(guò)G1/S檢測(cè)位點(diǎn),但并非所有進(jìn)入S期的細(xì)胞都能進(jìn)入正常的有絲分裂期。我們還發(fā)現(xiàn)Cdh1的缺失促進(jìn)肝實(shí)質(zhì)細(xì)胞過(guò)早的通過(guò)G1/S期的檢測(cè)位點(diǎn)主要是通過(guò)過(guò)度激活Ras信號(hào)通路來(lái)實(shí)現(xiàn)的。
[Abstract]:Cdc20 plays an important role in cell cycle regulation by ubiquitination and degradation of important cell cycle regulatory factors . Cdc20 plays an important role in the early stage of cell cycle . However , Cdh1 plays an important role in the early stage of mitosis and G1 phase .

The results showed that Cdh1 plays an important role not only in the transformation process of G1 / S phase , but also plays an important role in the life activities related to cell cycle .
In order to study the role of Cdh1 in the late and postnatal development of mice , Cdh1 conditional knockout mice were used as the research subjects .

In this study , it was found that Cdh1 ' s deletion would lead to the early entry of liver parenchyma cells into S phase , but the deletion of Cdh1 did not have a significant effect on the liver weight - weight ratio of mice .
