慢性鋁暴露對(duì)大鼠海馬LTP的影響及其機(jī)制研究

【摘要】: 目的: 應(yīng)用神經(jīng)電生理學(xué)、生物學(xué)等檢測(cè)方法,觀察慢性鋁暴露后大鼠學(xué)習(xí)記憶行為學(xué)、海馬長(zhǎng)時(shí)程增強(qiáng)(LTP)、細(xì)胞外信號(hào)調(diào)節(jié)激酶(ERK)蛋白以及ERK mRNA表達(dá)變化,旨在探討不同劑量鋁暴露對(duì)大鼠學(xué)習(xí)、記憶影響的作用機(jī)制,為中醫(yī)學(xué)“腦主神明”理論提供實(shí)驗(yàn)依據(jù)。 材料與方法: 1.實(shí)驗(yàn)動(dòng)物與分組選擇斷乳后30±5克Wistar雄性大鼠30只(由遼寧中醫(yī)藥大學(xué)實(shí)驗(yàn)動(dòng)物中心提供),30只大鼠隨機(jī)分為3組,每組10只大鼠。3組分別為:對(duì)照組(給予蒸餾水)、低劑量組(給予0.3% AlCl3水溶液,蒸餾水配制)、高劑量組(給予0.6% AlCl3水溶液,蒸餾水配制)。各組通過(guò)自由飲水?dāng)z入AICl3,連續(xù)服用AICl3染毒40天。動(dòng)物室溫度20-23℃,相對(duì)濕度50%-55%。 2.行為學(xué)觀察連續(xù)觀察5天各組大鼠圓形Morris水迷宮學(xué)習(xí)記憶行為學(xué)表現(xiàn)。 3.電生理觀察細(xì)胞外微電極記錄各組大鼠海馬DG-CA3途徑的LTP,記錄群體鋒電位(population spike, PS)。 4. RT-PCR檢測(cè)RT-PCR測(cè)定各組大鼠海馬ERK mRNA的表達(dá)。 5.蛋白檢測(cè)蛋白免疫印跡(Western-Blot)方法測(cè)定各組大鼠海馬ERK表達(dá)。 結(jié)果: 1.行為學(xué)表現(xiàn)與對(duì)照組比較,低劑量組、高劑量組水迷宮學(xué)習(xí)記憶的行為學(xué)表現(xiàn),大鼠找到平臺(tái)用時(shí)增長(zhǎng),有顯著差異(p0.05);高劑量組與低劑量組比較,大鼠找到平臺(tái)用時(shí)增長(zhǎng),有顯著差異(P0.05)。 2.電生理學(xué)改變與對(duì)照組比較,低劑量組、高劑量組海馬DG-CA3途徑的LTP,隨著鋁暴露劑量的增加,LTP增加的群鋒電位(PS)幅值比率逐漸下降,有顯著差異(p0.05)。 3.基因表達(dá)與對(duì)照組比較,低劑量組、高劑量組ERK mRNA表達(dá)降低,有顯著差異(p0.05)。 4.蛋白表達(dá)與對(duì)照組比較,低劑量組、高劑量組ERK Western-Blot表達(dá)降低,有顯著差異(p0.05)。 結(jié)論: 1.慢性鋁暴露,使大鼠記憶力減退,并呈劑量依賴(lài)性關(guān)系。 2.慢性鋁暴露,導(dǎo)致LTP增加的PS(群體鋒電位)幅值比率逐漸下降。 3.慢性鋁暴露使ERK的mRNA表達(dá)顯著降低。 4.慢性鋁暴露使ERK的蛋白表達(dá)顯著降低。
[Abstract]:Objective:The changes of learning and memory behavior, hippocampal long-term potentiation, extracellular signal regulated kinase (ERK) protein and ERK mRNA expression in rats after chronic aluminum exposure were observed by electrophysiological and biological methods.The purpose of this study was to explore the mechanism of the effects of different doses of aluminum exposure on learning and memory in rats, and to provide experimental basis for the theory of "brain dominating Shenming" in traditional Chinese medicine.Materials and methods:1.The experimental animals were divided into three groups: 30 鹵5 g Wistar male rats after weaning (30 rats provided by the Experimental Animal Center of Liaoning University of traditional Chinese Medicine) were randomly divided into 3 groups.Each group of 10 rats were divided into three groups: control group (treated with distilled water), low dose group (treated with 0.3% AlCl3 solution, distilled water), high dose group (0.6% AlCl3 solution, distilled water).Each group received AICL 3 through free drinking water for 40 days.The temperature of the animal room is 20-23 鈩,
