寵物犬藍(lán)氏賈第鞭毛蟲和隱孢子蟲感染的分子檢測

【摘要】:目的了解皖浙兩省部分城鎮(zhèn)地區(qū)寵物犬藍(lán)氏賈第鞭毛蟲和隱孢子蟲感染情況,及聚集體類型或蟲種。方法在安徽多地和浙江杭州共采集315份新鮮寵物犬糞樣,分別采用基于藍(lán)氏賈第鞭毛蟲谷氨酸脫氫酶(GDH)基因的半巢式PCR和隱孢子蟲核糖體小亞基(SSU r RNA)基因的巢式PCR方法對(duì)所有糞樣進(jìn)行擴(kuò)增,并對(duì)獲得的陽性擴(kuò)增產(chǎn)物進(jìn)行測序和生物信息學(xué)分析,確定藍(lán)氏賈第鞭毛蟲的聚集體和隱孢子蟲的蟲種類型。結(jié)果 315份犬糞樣中,藍(lán)氏賈第鞭毛蟲和隱孢子蟲的陽性率分別為3.2%(10/315)和1.6%(5/315)。幼齡犬(≤12個(gè)月)藍(lán)氏賈第鞭毛蟲陽性率(17.8%)和隱孢子蟲陽性率(11.1%)均顯著高于成年犬(0.7%和0)(P0.05)。雌性犬和雄性犬藍(lán)氏賈第鞭毛蟲和隱孢子蟲陽性率差異均無統(tǒng)計(jì)學(xué)意義(P0.05)。發(fā)現(xiàn)藍(lán)氏賈第鞭毛蟲兩種聚集體類型,即聚集體B(n=6)和聚集體D(n=4)。從5份陽性犬糞中分離的隱孢子蟲經(jīng)巢式PCR SSU r RNA基因檢測,均為犬隱孢子蟲(Cryptosporidium canis)。結(jié)論皖浙兩省部分地區(qū)寵物犬藍(lán)氏賈第鞭毛蟲和犬隱孢子蟲的陽性率分別為3.2%和1.6%。發(fā)現(xiàn)的藍(lán)氏賈第鞭毛蟲聚集體類型為聚集體B和聚集體D。
[Abstract]:Objective to investigate the infection of Giardia lamblia and Cryptosporidium in pet dogs in some cities and towns of Anhui and Zhejiang provinces. Methods 315 fresh dog feces were collected in Anhui Province and Hangzhou Zhejiang Province. All fecal samples were amplified by nested PCR and Cryptosporidium ribosomal small subunit SSU r RNAs, respectively, based on the GDH gene of Giardia lamblia. The positive amplified products were sequenced and bioinformatics analysis to determine the aggregates of Giardia lamblia and the species types of Cryptosporidium.
