雷公藤紅素降低糖尿病小鼠血糖的機制研究

【摘要】:目的:雷公藤紅素(Celastrol)是一種三萜類化合物,最早從南蛇藤莖中分離得到,具有抗炎,抗癌等多種活性,臨床上主要用于治療類風(fēng)濕關(guān)節(jié)炎,皮膚病和腎臟疾病。我們的前期研究表明在適當(dāng)?shù)膭┝肯拢?00μg/kg),雷公藤紅素具有很明顯的降低由鏈脈佐菌素(STZ,50mg/kg)所誘導(dǎo)產(chǎn)生糖尿病小鼠血糖的作用。為了進(jìn)一步探討雷公藤紅素的作用機理,本文通過建立細(xì)胞和動物模型,研究了雷公藤紅素對其功能的影響。 方法:通過炎癥因子(IL-1,IL-6)將β-胰島細(xì)胞(INS-1)感染成炎癥狀態(tài)后,加入雷公藤紅素(100nM-300nM)治療處理,觀察細(xì)胞狀態(tài)(Immunofluorescence technique)的變化,并測定其功能(KSIS),細(xì)胞凋亡水平(FCM)。 建立C57BL/6J小鼠的糖尿病模型,對其體重(BW),隨機血糖(GLU),24小時尿糖(UGE),尿蛋白(UP),腎功能(Renal Function)等指標(biāo)進(jìn)行測定,并提取腎小管上皮細(xì)胞總RNA,檢測鈉葡萄糖共轉(zhuǎn)移蛋白(SGLT)表達(dá)的變化。 結(jié)果:INS-1細(xì)胞通過雷公藤紅素處理24小時之后,與對照組相比,β-胰島細(xì)胞狀態(tài)和功能明顯好轉(zhuǎn),細(xì)胞凋亡得到了抑制。糖尿病小鼠經(jīng)雷公藤紅素治療后,血糖明顯降低,24小時尿糖增多,尿蛋白增加,,腎小球橫截面積增加,SGLT的表達(dá)明顯降低。 結(jié)論:雷公藤紅素對炎癥狀態(tài)下β-胰島細(xì)胞具有一定的保護作用,通過對鈉葡萄糖共轉(zhuǎn)運體的抑制,減少了葡萄糖的重吸收,從而降低糖尿病小鼠的血糖。
[Abstract]:Objective: Tripterygium wilfordii Celastrola is a triterpenoid compound, which was first isolated from the stem of Sagitaria sinensis. It has many anti-inflammatory and anticancer activities, and is mainly used in the treatment of rheumatoid arthritis. Skin diseases and kidney diseases. Our previous studies have shown that tripterygium at an appropriate dose of 500 渭 g / kg, tripteryline can significantly reduce the effect of streptozotocin (STZ) 50 mg / kg on the production of blood glucose in diabetic mice. The mechanism of the action of tripsin, The effects of tripterine on the function of tripterygium wilfordii were studied by establishing cell and animal models. Methods: 尾 -islet cells (INS-1) were infected into inflammatory state by inflammatory cytokine IL-1- (IL-6), then treated with Tripterygium wilfordii 100nM-300nM), the changes of immunofluorescence technique were observed, and the function of KSISI and the level of apoptosis were measured. The expression of SGLT was significantly decreased after 24 hours of blood glucose decreased, urine protein increased and glomerular cross-sectional area increased. Conclusion: Tripterygium wilfordii has a protective effect on 尾 -islet cells in inflammatory state. By inhibiting sodium and glucose co-transporter, tripterygium can reduce the reabsorption of glucose and decrease the blood glucose in diabetic mice.
