大鼠成纖維細胞生長因子及相關(guān)基因的表達與鼠齡關(guān)系的初步研究

【摘要】: 目的: 通過對不同鼠齡大鼠在同樣條件下牽引成骨效果的比較,研究成纖維細胞生長因子及相關(guān)基因在成骨局部的表達水平與年齡的關(guān)系,及其在大鼠脛骨牽引成骨過程中對新骨形成的影響。 方法: 鼠齡分別為3個月和12個月的健康雄性SD大鼠各10只,分為青年組(n=10)和老年組(n=10)。所有大鼠接受左脛骨中上段低能截骨,安置自制微型體外延長固定架。術(shù)后第二天起開始每天進行脛骨延長,速率為0.2mm/次,2次/天,共14天。術(shù)后第15天,處死大鼠,采集脛骨標本,分別進行影像學(xué)及組織學(xué)檢查和RT-PCR (Reverse Transcription-Polymerase Chain Reaction)檢測,以測定脛骨延長段骨痂中新生骨量及成纖維細胞生長因子及相關(guān)基因的表達。所有數(shù)據(jù)采用t檢驗,進行統(tǒng)計學(xué)處理。 結(jié)果: (1)青年組大鼠骨牽引區(qū)有大量骨痂,新生成骨面積和新生骨密度均顯著高于老年組大鼠; (2)成纖維細胞生長因子及相關(guān)基因在老年組大鼠的表達水平明顯低于青年組。 結(jié)論: 老年大鼠骨形成障礙可能與局部生長因子表達降低,從而導(dǎo)致成骨細胞功能低下密切相關(guān)。
[Abstract]:Objective:. By comparing the effects of distraction osteogenesis of rats of different ages under the same conditions, the relationship between the expression level of fibroblast growth factor and related genes in osteoblasts was studied. And its effect on the formation of new bone during distraction osteogenesis of rat tibia. Methods:. Ten male Sprague-Dawley rats aged 3 months and 12 months old were divided into two groups: the young group (n = 10) and the old group (n = 10). All the rats received a left middle and upper tibia osteotomy. From the second day after operation, the tibia was lengthened daily at the rate of 0.2mm/ twice a day for 14 days. On the 15th day after operation, the rats were sacrificed and the tibia specimens were collected. Imaging and histological examination and RT-PCR reverse Transcription-Polymerase Chain reaction were performed to determine the amount of new bone and the expression of fibroblast growth factor and related genes in callus of tibial lengthened segment. Results:. 1) there was a large amount of callus in the bone traction area of the young rats, and the area of new osteogenesis and the bone mineral density of the young rats were significantly higher than those of the elderly rats. (2) the expression of fibroblast growth factor and related genes in the aged group was significantly lower than that in the young group. Conclusion:. The dysfunction of bone formation in aged rats may be related to the decreased expression of local growth factor, which leads to the low function of osteoblasts.
