【摘要】: 乳酸作為糖代謝的中間產(chǎn)物在外周神經(jīng)系統(tǒng)(聯(lián)系中樞神經(jīng)系統(tǒng)與全身各器官的神經(jīng),包括腦神經(jīng)和脊神經(jīng))的作用已經(jīng)得到非常廣泛研究。劇烈運(yùn)動時(shí),外周乳酸濃度顯著增高,乳酸積累被認(rèn)為是產(chǎn)生疲勞的機(jī)制之一。另外,近年來的一些研究發(fā)現(xiàn),運(yùn)動中體內(nèi)氨的積累與疲勞的產(chǎn)生也密切相關(guān)。SO_2是常見的大氣污染物,尤其在一些工業(yè)較發(fā)達(dá)、人口較密集或燃用高硫煤較多的城市或地區(qū)SO_2超標(biāo)非常嚴(yán)重。因此,SO_2對人體健康影響的研究日益受到社會的關(guān)注。 為了從信號轉(zhuǎn)導(dǎo)角度探討SO_2對大鼠的降壓作用機(jī)理、SO_2對血管舒張作用的機(jī)理,本實(shí)驗(yàn)以Wistar大鼠為實(shí)驗(yàn)對象,測定SO_2對體外培養(yǎng)平滑肌細(xì)胞、內(nèi)皮細(xì)胞的影響。另外,本實(shí)驗(yàn)初步研究了乳酸以及氨對Wistar大鼠胸主動脈血管的舒張作用。 1、乳酸和氨對血管舒張的影響及與SO_2的聯(lián)合作用 研究了乳酸和氨對血管舒張的影響以及其與其他藥物的聯(lián)合作用。采用文獻(xiàn)的方法制備大鼠胸主動脈環(huán),Medlab-U/4C501生物信號采集處理系統(tǒng)測定血管環(huán)張力,分別加入不同濃度的乳酸、氨水以及其他藥物。結(jié)果:①在10~(-6)mol·L~(-1)NE預(yù)收縮后,加入乳酸(0.5、1、5、10、20、100mmol·L~(-1))后血管環(huán)張力逐漸減小,表現(xiàn)為舒張作用,且與乳酸的濃度呈劑量依賴性。分別加入10μmol·L~(-1)、100μmol·L~(-1)SO_2;10μmol·L~(-1)、100μmol·L~(-1)亞硫酸鹽;3nmol·L~(-1)、5nmol·L~(-1)硝普鈉,并同時(shí)加入不同濃度的乳酸后血管環(huán)張力逐漸減小,均表現(xiàn)為舒張作用。但其舒張比小于乳酸或其他藥物單獨(dú)作用后舒張比之和。②氨(0.3-20mmol·L~(-1))對大鼠主動脈環(huán)有濃度依賴性的舒張作用;低濃度氨(0.3-1.0mmol·L~(-1))對血管的舒張作用無明顯影響,而高濃度氨(2.0-20 mmol·L~(-1))的影響比較明顯。 2、SO_2對血管細(xì)胞產(chǎn)生Ca~(2+)濃度的影響 研究了不同濃度SO_2對大鼠胸主動脈平滑肌、內(nèi)皮細(xì)胞鈣離子通道的作用機(jī)制。采用剝離內(nèi)膜塊的組織貼塊法分離大鼠主動脈EC及SMC,分別以5、15、25、50、100、500、1000、2000μmol·L~(-1) SO_2水溶液處理大鼠胸主動脈平滑肌細(xì)胞,以200、1000、2000μmol·L~(-1) SO_2水溶液處理大鼠胸主動脈內(nèi)皮細(xì)胞,用激光共聚焦顯微鏡掃描測定細(xì)胞中的自由鈣離子濃度。結(jié)果顯示:①累計(jì)加入濃度為5、15、25、50、100、500、1000、2000μmol·L~(-1)SO_2水溶液后,與對照組相比,平滑肌細(xì)胞內(nèi)的鈣離子熒光強(qiáng)度分別下降到98.58%,90.10%,85.09%,79.56%,73.13%,70.21%,11.35%和8.72%。用NE(1μmol·L~(-1))預(yù)處理血管平滑肌細(xì)胞后,顯著削弱了高濃度SO_2(1000μmol·L~(-1))降低[Ca~(2+)]_i的作用,但卻沒有削弱低濃度SO_2(10、50μmol·L~(-1))降低[Ca~(2+)]_i的作用。 ②累計(jì)加入濃度為200、1000、2000μmol·L~(-1)SO_2水溶液后,與對照組相比,內(nèi)皮細(xì)胞內(nèi)的鈣離子熒光強(qiáng)度分別下降到90.90%,68.97%,7.06%。
[Abstract]:The role of lactic acid as an intermediate of glucose metabolism in the peripheral nervous system, which connects the central nervous system with all the organs of the body, including the brain and spinal nerves, has been extensively studied. The accumulation of lactic acid is considered to be one of the mechanisms of fatigue. In addition, some recent studies have found that ammonia accumulation in exercise is closely related to fatigue. Especially in some cities or regions where the industry is more developed, the population is more dense or the high sulfur coal is burned, the SO_2 exceeds the standard very seriously. Therefore, the study of the effect of so _ 2 on human health has been paid more and more attention by the society. In order to investigate the mechanism of antihypertensive effect of SO_2 on rats and the mechanism of vasodilation induced by SO-2 from the point of view of signal transduction, the effects of SO_2 on cultured smooth muscle cells and endothelial cells were measured in Wistar rats. The effects of lactic acid and ammonia on thoracic aortic vasodilation in Wistar rats were studied. 1. Effects of lactic acid and ammonia on vasodilation and their combined effects with SO_2.
