針刺、艾灸對(duì)AA模型大鼠痛閾及“足三里”穴位局部組織液中鉀離子的不同影響

【摘要】:目的:研究針灸對(duì)AA模型大鼠痛閾以及“足三里”穴位局部組織液中鉀離子濃度的影響,以期從穴位局部相關(guān)物質(zhì)變化這個(gè)角度出發(fā),探索針刺與艾灸對(duì)痛閾與穴位局部組織液中鉀離子濃度的不同影響,同時(shí)也為后期研究針灸穴位局部機(jī)制提供新的實(shí)驗(yàn)依據(jù)。 方法:以AA模型大鼠為炎性疼痛的研究載體,一批大鼠針灸“足三里”30min,用Von Frey測(cè)痛儀檢測(cè)針灸前,針灸中及針灸后2h痛閾變化,另一批大鼠針灸“足三里”30min,同時(shí)采用微透析儀于“足三里”穴位局部收集針灸前,針灸中及針灸后2h組織液,每30min收集一個(gè)樣品,將所得樣品即時(shí)用電解質(zhì)分析儀檢測(cè)胞外鉀離子濃度。統(tǒng)計(jì)分析組內(nèi)比較用重復(fù)測(cè)量方差分析法,組間比較采用T檢驗(yàn)法,P0.05表示有統(tǒng)計(jì)學(xué)差異。 結(jié)果:針刺及艾灸后,AA模型大鼠痛閾高于針灸前(p0.05),“足三里”穴位局部組織液中鉀離子濃度也高于針灸前(p0.05),空針組痛閾高于空灸組(p0.05),模灸組痛閾高于模針組(p0.05),空灸組穴位局部胞外鉀離子濃度高于空針組(p0.05),模灸組穴位局部胞外鉀離子濃度高于模針組(p0.05)。結(jié)論:針灸能提高AA模型大鼠痛閾;針刺和艾灸對(duì)痛閾和穴位局部胞外鉀離子濃度的影響有細(xì)微區(qū)別。對(duì)正常大鼠,針刺組痛閾高于艾灸組,而艾灸對(duì)穴位局部胞外鉀離子的升高作用,高峰到達(dá)所用時(shí)間短。對(duì)AA模型大鼠,艾灸組痛閾高于針刺組,并且艾灸后痛閾高峰維持時(shí)間較長(zhǎng),艾灸對(duì)于胞外鉀離子的升高作用,維持高濃度時(shí)間也較長(zhǎng)。
[Abstract]:Objective: to study the effect of acupuncture and moxibustion on pain threshold and potassium ion concentration in local tissue fluid of "Zusanli" acupoint in AA model rats. To explore the different effects of acupuncture and moxibustion on the concentration of potassium ions in pain threshold and local tissue fluid of acupoints, and to provide a new experimental basis for the later study of local mechanism of acupuncture and moxibustion. Methods: AA model rats were used as the research carrier of inflammatory pain. A batch of rats were treated with acupuncture and moxibustion "Zusanli" for 30 minutes. The pain threshold before acupuncture, during acupuncture and 2 hours after acupuncture were detected by Von Frey. Another group of rats received acupuncture and moxibustion "Zusanli" for 30 mins. At the same time, a sample was collected every 30 minutes before acupuncture, 2 hours after acupuncture and 2 hours after acupuncture and moxibustion, using microdialysis apparatus at Zusanli acupoint. The concentration of extracellular potassium ion was detected by electrolyte analyzer immediately. The statistical analysis group was compared with repeated measurement variance analysis. Results: the pain threshold of AA model rats after acupuncture and moxibustion was higher than that before acupuncture, the concentration of potassium ion in local tissue fluid of Zusanli acupoint was also higher than that before acupuncture, the pain threshold of empty acupuncture group was higher than that of empty moxibustion group, the pain threshold of model moxibustion group was higher than that of model acupuncture group, and the pain threshold of model moxibustion group was higher than that of model acupuncture group. The concentration of extracellular potassium ion at acupoints in moxibustion group was higher than that in empty acupuncture group, and the concentration of extracellular potassium ion in moxibustion group was higher than that in model acupuncture group. Conclusion: acupuncture can increase pain threshold of AA model rats. The effects of acupuncture and moxibustion on pain threshold and local extracellular potassium concentration at acupoints were slightly different. For normal rats, the pain threshold of acupuncture group was higher than that of moxibustion group, while moxibustion could increase the level of extracellular potassium at acupoints. For AA model rats, the pain threshold of moxibustion group was higher than that of acupuncture group, and the time of maintaining the peak of pain threshold after moxibustion was longer, and the effect of moxibustion on the increase of extracellular potassium ion and the duration of high concentration of moxibustion were also longer.
