【摘要】：目的:探討氧化低密度脂蛋白(oxidized low density lipoprotein,ox-LDL)對巨噬細胞源性泡沫細胞吞噬功能和炎癥相關(guān)因子分泌功能的影響。方法:利用佛波酯(phorbol ester,PMA)誘導(dǎo)THP-1細胞分化形成巨噬細胞,之后采用ox-LDL處理48小時后,誘導(dǎo)其形成泡沫細胞。利用中性紅吞噬實驗,分析泡沫細胞形成前后吞噬功能的變化;通過ELISA法,檢測細胞培養(yǎng)上清中腫瘤壞死因子α(tumor necrosis factorα,TNF-α)含量,觀察ox-LDL對THP-1巨噬細胞功能的影響。結(jié)果:細胞形態(tài)學(xué)結(jié)果表明,我們成功利用ox-LDL誘導(dǎo)THP-1巨噬細胞形成泡沫細胞;進一步發(fā)現(xiàn)ox-LDL誘導(dǎo)THP-1巨噬細胞表面的清道夫受體CD36表達升高,并促進細胞吞噬功能增加,進一步促進細胞內(nèi)膽固醇含量顯著升高(P0.05);同時,ox-LDL能夠刺激巨噬細胞大量分泌TNF-α(P0.05)。結(jié)論:ox-LDL通過增強清道夫受體CD36表達,提高巨噬細胞的吞噬功能,引起大量膽固醇聚集,產(chǎn)生細胞毒性損傷,并促進TNF-α炎性因子的大量分泌。
[Abstract]:Objective: To investigate the effects of oxidized low density lipoprotein (oxidized low density lipoprotein, ox-LDL) related factors affecting phagocytic function and inflammatory secretion of macrophage derived foam cells. Methods: using phorbol-12-myristate-13-acetate (phorbol ester PMA) THP-1 cell differentiation induced by formation of macrophage cells, after treated with ox-LDL for 48 hours, induced the formation of foam cells. Using neutral red phagocytosis test, analysis of the formation of foam cells before and after phagocytosis; by ELISA method, the detection of tumor necrosis factor alpha cell culture supernatant (tumor necrosis factor TNF- alpha, alpha) content, the effect of ox-LDL on function of THP-1 macrophage cell morphology. Results: the results showed that we successfully induced by ox-LDL THP-1 macrophage foam cell formation; we found that ox-LDL induced THP-1 macrophage scavenger receptor CD36 expression increased, and promote phagocytosis To further promote the function to increase the intracellular cholesterol content was significantly increased (P0.05); at the same time, ox-LDL can stimulate the secretion of macrophage TNF- alpha (P0.05). Conclusion: ox-LDL can enhance the expression of scavenger receptor CD36, improve the phagocytic function of macrophage, causing a lot of cholesterol accumulation, produce cytotoxic injury and promote the secretion of inflammatory TNF- alpha factor.
