【摘要】： 本研究利用分子生物學(xué)、免疫學(xué)技術(shù)構(gòu)建重組基因疫苗pcDNA3- MAGE-1和pcDNA3-hIL-18(小鼠pcDNA3-mIL-18由本實(shí)驗(yàn)室保存);探討聯(lián)合應(yīng)用基因疫苗體內(nèi)、外抗肝癌免疫治療的作用。基因疫苗分組免疫實(shí)驗(yàn)小鼠,在體外驗(yàn)證疫苗免疫后的小鼠脾細(xì)胞對(duì)MAGE-1陽性和陰性肝癌細(xì)胞株的殺傷作用;同時(shí)我們探討不同劑量pcIL-18在與pcMAGE-1聯(lián)合應(yīng)用時(shí)達(dá)到最佳殺傷效應(yīng)的免疫劑量。將MAGE-1轉(zhuǎn)染至小鼠肝癌Hepa1-6細(xì)胞,獲得穩(wěn)定表達(dá)MAGE-1基因的小鼠肝癌Hepa1-6-MAGE-1細(xì)胞系,為本研究以及進(jìn)一步研究MAGE-1在腫瘤生物治療中的應(yīng)用奠定基礎(chǔ)。體內(nèi)抗肝癌的實(shí)驗(yàn)研究,各實(shí)驗(yàn)組小鼠先皮下注射Hepa1-6-MAGE-1細(xì)胞和Hepa1-6細(xì)胞再接種基因疫苗。觀察不同基因疫苗對(duì)MAGE-1(+)和MAGE-1(-)荷瘤小鼠腫瘤生長(zhǎng)和生存時(shí)間的影響。本研究結(jié)果表明MAGE-1與較小劑量IL-18的聯(lián)合應(yīng)用具有明顯的抗肝癌免疫治療作用。
[Abstract]:The purpose of this study was to construct recombinant gene vaccine pcDNA3- MAGE-1 and pcDNA3-hIL-18 by molecular biology and immunology. The killing effect of spleen cells of mice immunized with vaccine on MAGE-1 positive and negative hepatoma cell lines was verified in vitro. At the same time, we studied the optimal dose of pcIL-18 in combination with pcMAGE-1. MAGE-1 was transfected into mouse hepatoma Hepa1-6 cells to obtain murine hepatocarcinoma Hepa1-6-MAGE-1 cell line which expressed MAGE-1 gene stably. For this study and further study of the application of MAGE-1 in tumor biotherapy. Experimental study of anti-hepatoma in vivo, Mice in each experimental group were injected subcutaneously with Hepa1-6-MAGE-1 cells and Hepa1-6 cells and then inoculated with gene vaccine. The effects of different gene vaccines on tumor growth and survival time of mice bearing MAGE-1 () and MAGE-1-) were observed. The combined use of the drug has obvious anti-hepatoma immunotherapy effect.
