改良的光化學(xué)法血栓模型的建立及消栓通絡(luò)湯對(duì)血栓形成的影響

【摘要】: 目的:建立一種以光化學(xué)反應(yīng)為基礎(chǔ)的改良的光化學(xué)法動(dòng)物血栓模型;基于已經(jīng)建立起來(lái)的改良的光化學(xué)法動(dòng)物血栓模型來(lái)探討消栓通絡(luò)湯對(duì)改良的光化學(xué)法動(dòng)物血栓模型的作用;對(duì)治療血栓性疾病的臨床驗(yàn)方消栓通絡(luò)湯的處方結(jié)構(gòu)進(jìn)行優(yōu)化研究。方法:小鼠經(jīng)尾靜脈注射給予孟加拉紅,將小鼠耳部平鋪于生物機(jī)能實(shí)驗(yàn)系統(tǒng)和微循環(huán)觀測(cè)系統(tǒng)的顯微鏡下,用波長(zhǎng)為490nm的冷光源照射小鼠耳廓目標(biāo)血管,觀察并記錄小鼠耳廓目標(biāo)血管的血流狀態(tài),以不同時(shí)間點(diǎn)血流速度及血流完全停止的時(shí)間作為評(píng)價(jià)血栓形成的指標(biāo);通過(guò)對(duì)小鼠一個(gè)月的灌胃給藥后,用改良的光化學(xué)法對(duì)小鼠進(jìn)行造模,觀察消栓通絡(luò)湯組及阿司匹林組之間小鼠耳廓血管不同時(shí)間點(diǎn)的血流速度變化、血流停止時(shí)間、血小板數(shù)變化的差異作為評(píng)價(jià)消栓通絡(luò)湯對(duì)改良的光化學(xué)法動(dòng)物血栓模型影響的指標(biāo);運(yùn)用均勻設(shè)計(jì)改良法,以小鼠不同時(shí)間點(diǎn)的血流速度變化、血流停止時(shí)間、血小板數(shù)變化的藥效學(xué)指標(biāo)為評(píng)價(jià)指標(biāo),進(jìn)行處方最佳配伍和最佳劑量配比的比較研究。結(jié)果:孟加拉紅組在冷光源照射5min時(shí),小鼠耳廓血管血流速度明顯減慢,照射到25min時(shí),血管血流完全停止;消栓通絡(luò)湯組冷光源照射5min后,血流速度明顯減慢,照射到25min時(shí),高劑量組、中劑量組血流未停止,低劑量組的血流完全停止,隨著劑量的增加,對(duì)血小板的聚集的抑制作用增強(qiáng);以均勻設(shè)計(jì)法得出了消栓通絡(luò)湯最佳配伍及最佳劑量配比的理論值,并經(jīng)驗(yàn)證確認(rèn),提取物10(由水蛭3g,當(dāng)歸27g,地龍24g,川芎21g,懷牛膝15g,石菖蒲9g組成)和理論4組(由水蛭3g,當(dāng)歸30g,地龍30g,川芎25g,懷牛膝20g組成)藥效最佳,因理論4組藥味少1種,故可推薦用于后續(xù)新藥開(kāi)發(fā)研究。結(jié)論:490nm冷光源照射已經(jīng)注射孟加拉紅的小鼠耳廓血管,能夠造成小鼠耳廓血管的血流速度減慢,最終血流停止,可以建立以光化學(xué)反應(yīng)為基礎(chǔ)的動(dòng)物血栓模型;消栓通絡(luò)湯延緩改良的光化學(xué)動(dòng)物血栓模型的血流速度減慢的速度、延長(zhǎng)血流停止時(shí)間、抑制血小板聚集,具有一定的抗血栓形成作用;本實(shí)驗(yàn)研究方法將拆方分析與最佳劑量配比研究結(jié)合起來(lái),運(yùn)用于處方優(yōu)化具有切實(shí)可行性。
[Abstract]:Objective: to establish a modified photochemical animal thrombus model based on photochemical reaction. Based on the established animal thrombus model, the effect of Xiaoshuantongluo decoction on the modified photochemical animal thrombus model was studied. The prescription structure of Xiaoshuantongluo decoction, a clinical prescription for treating thrombotic diseases, was optimized. Methods: the mice were injected with Bengal red through tail vein. The ears of mice were laminated under the microscope of biological function test system and microcirculation observation system. The target blood vessels of mouse auricle were irradiated with a cold light source of 490nm, and the blood flow status of the target vessels of mouse auricle was observed and recorded. The blood flow velocity and the time when the blood flow stopped completely at different time points were taken as the indexes to evaluate thrombosis, and the mice were modeled by modified photochemical method after one month of intragastric administration. To observe the changes of blood flow velocity and blood flow stop time of auricle blood vessel of mice at different time points between the group of Xiao Shuan Tongluo decoction and the group of aspirin. The difference of platelet number was used as an index to evaluate the effect of Xiaoxuan Tongluo decoction on the modified photochemical animal thrombus model, and the uniform design method was used to change the blood flow velocity and stop time of blood flow at different time points in mice. The pharmacodynamics index of platelet number change was used as the evaluation index. The best prescription compatibility and the best dose ratio were compared. Results: the blood flow velocity of auricle blood vessel of mice in Bengal red group was significantly decreased when exposed to cold light source for 5 min, and at 25 min after exposure to Bengal red group. The blood flow of the blood vessel stopped completely in the Xiaoshuantongluo decoction group, and the blood flow velocity of the cold light source in the Xiaoshuantongluo decoction group was obviously slowed down after 5 min irradiation. At 25 min, the blood flow in the high dose group and the middle dose group did not stop, and the blood flow in the low dose group stopped completely with the increase of the dose. The inhibitory effect on platelet aggregation was enhanced, and the theoretical values of the best compatibility and the best dosage ratio of Xiaoxuan Tongluo decoction were obtained by uniform design, and confirmed by verification, Extract 10 (composed of Hirudo 3g, Angelica 27g, Dilong 24g, Ligusticum chuanxiong 21g, Radix bidentata 15g, Acorus calamus 9g) and theoretical group 4 (composed of leeches 3g, angelica sinensis 30g, Dilong 30g, Chuanxiong 25g, Achyranthes bidentata 20g) had the best effect, because the theoretical 4 groups had one less flavor, Conclusion irradiation of auricle vessels injected with Bengal red can slow down the blood flow velocity of the auricle vessels in mice and eventually stop the blood flow. The animal thrombus model based on photochemical reaction can be established, and Xiaoshuantongluo decoction can slow down the blood flow speed, prolong the blood flow stop time and inhibit platelet aggregation in the modified photochemical animal thrombus model. The experimental method combines the disassembly analysis with the optimal dose ratio study, and it is feasible to apply to the optimization of prescription.
