異種關(guān)節(jié)軟骨與MSCs體外共同培養(yǎng)的實(shí)驗(yàn)研究

【摘要】:目的提供一種新的種子細(xì)胞獲得方法,解決軟骨缺損修復(fù)過(guò)程中軟骨數(shù)量不足的困難。方法人骨髓間充質(zhì)干細(xì)胞體外擴(kuò)增誘導(dǎo)分化后與兔的關(guān)節(jié)軟骨細(xì)胞按不同比例共同培養(yǎng)利用real-time PCR技術(shù)分析細(xì)胞表型的表達(dá)情況。結(jié)果實(shí)驗(yàn)組2種細(xì)胞共培養(yǎng)4周后,混合培養(yǎng)顯示正反饋調(diào)節(jié)軟骨細(xì)胞的增殖,軟骨細(xì)胞外基質(zhì)、SOX9、Ⅱ型膠原基因表達(dá)增加,軟骨細(xì)胞生成明顯增加。結(jié)論間充質(zhì)干細(xì)胞以旁分泌或自分泌的方式調(diào)節(jié)間充質(zhì)干細(xì)胞向軟骨細(xì)胞的轉(zhuǎn)化。
[Abstract]:Objective to provide a new method for obtaining seed cells. Methods Human bone marrow mesenchymal stem cells were cultured with rabbit articular chondrocytes in different proportions to analyze the cell surface by real-time PCR. Results the two kinds of cells were co-cultured for 4 weeks in the experimental group.
