大鼠自體外周血內(nèi)皮前體細(xì)胞培養(yǎng)的實(shí)驗(yàn)研究

【摘要】: 目的探討自大鼠外周血獲取內(nèi)皮前體細(xì)胞(endothelial progenitorcells EPCS)的方法,為干細(xì)胞移植治療缺血性心肌損傷提供新的無創(chuàng)性的細(xì)胞來源。 方法抽取經(jīng)連續(xù)皮下注射大鼠粒細(xì)胞集落刺激因子(Rat GranulocyteColony Stimulating Factor rG-CSF)七天的成年雄性Sprague-Dawley(SD)大鼠外周血,應(yīng)用聚蔗糖(Ficoll 400)密度梯度離心法分離提取大鼠外周血中的單個(gè)核細(xì)胞,然后以20%低糖IMDM、胎牛血清、馬血清、堿性成纖維細(xì)胞生長因子(basic fibroblast growth factor bFGF)和血管內(nèi)皮生長因子(vascular endothelial growth factor VEGF)等混合而成的完全培養(yǎng)液,進(jìn)行體外培養(yǎng)擴(kuò)增,培養(yǎng)所得細(xì)胞經(jīng)CD31、CD34、Flk-1和vW F免疫熒光染色并檢測其結(jié)合UEA-1、攝取ac-LDL的能力。 結(jié)果培養(yǎng)所得細(xì)胞經(jīng)CD31、CD34、Flk-1和vW F免疫熒光染色,并測定其結(jié)合UEA-1、攝取ac-LDL的能力,皆為陽性,證實(shí)為內(nèi)皮前體細(xì)胞(EPC)。 結(jié)論從粒細(xì)胞動(dòng)員后的大鼠外周血中獲取單個(gè)核細(xì)胞,經(jīng)體外培養(yǎng)誘導(dǎo),成功獲得內(nèi)皮前體細(xì)胞(EPC),內(nèi)皮前體細(xì)胞(EPC)有望成為移植治療缺血性心肌損傷的新的無創(chuàng)性細(xì)胞來源。
[Abstract]:Objective to investigate the method of acquiring endothelial progenitor cells (progenitorcells) from peripheral blood of rats, and to provide a new noninvasive cell source for stem cell transplantation in the treatment of ischemic myocardial injury. Methods the peripheral blood of adult male Sprague-Dawley SD rats injected subcutaneously with rat GranulocyteColony Stimulating Factor rG-CSF for 7 days was isolated and extracted by density gradient centrifugation with polysucrose Ficoll 400 (Ficoll 400), and the mononuclear cells were isolated from the peripheral blood of adult male Sprague-Dawley rats by density gradient centrifugation. Then, 20% low glucose IMDM, fetal bovine serum, horse serum, basic fibroblast growth factor basic fibroblast growth factor bFGFand vascular endothelial growth factor VEGF were mixed and cultured in vitro. The cultured cells were stained with CD31, CD34, Flk-1 and VWF, and the ability of UEA-1 to absorb ac-LDL was detected. Results the cultured cells were stained with CD31T CD34Flk-1 and VWF, and the ability of uptake of ac-LDL was positive, which was confirmed to be endothelial progenitor cells. Conclusion after granulocyte mobilization, mononuclear cells were obtained from peripheral blood of rats. After cultured and induced in vitro, endothelial progenitor cells (EPCs) were successfully obtained. EPCs of endothelial progenitor cells (EPCs) are expected to become a new noninvasive cell source for the treatment of ischemic myocardial injury.
