堿性成纖維生長因子對脂肪干細(xì)胞分化為心肌細(xì)胞的影響

【摘要】:【目的】探討堿性成纖維生長因子(b FGF)對脂肪干細(xì)胞(ADSC)分化為心肌細(xì)胞的影響!痉椒ā繌腟D大鼠中提取、分離、培養(yǎng)、鑒定ADSC后,評價ADSC分化能力。將成功建立心肌梗死(MI)模型的60只大鼠隨機(jī)分4組(n=15/組):PBS組、b FGF組、ADSC組及b FGF+ADSC組。建立心梗1周后分別注射PBS、b FGF、ADSC及b FGF+ADSC,4周后用心臟超聲測量心梗大鼠的左室射血分?jǐn)?shù)(EF),取大鼠心臟檢測左室梗死面積、微血管密度(MVD)及ADSC分化。【結(jié)果】成功分離ADSC,流式細(xì)胞分析顯示大多數(shù)ADSC表達(dá)CD29和CD90,不表達(dá)CD34和CD45。ADSC能被誘導(dǎo)分化成成脂、成骨細(xì)胞。b FGF+ADSC組及ADSC組的EF高于PBS組和b FGF組(P0.01),b FGF+ADSC組的EF高于ADSC組(P0.01),PBS組和b FGF組的EF差別無統(tǒng)計學(xué)意義(P=0.33)。b FGF+ADSC組及ADSC組的梗死面積小于PBS組和b FGF組(P0.01),b FGF+ADSC組的梗死面積小于ADSC組(P0.01),PBS組和b FGF組的梗死面積差別無統(tǒng)計學(xué)意義(P=0.28)。四組中b FGF+ADSC組的MVD最大(P0.01)。ADSC可分化為c Tn T陽性細(xì)胞、SMA陽性細(xì)胞及fⅧ陽性細(xì)胞!窘Y(jié)論】b FGF可促進(jìn)ADSC分化成心肌細(xì)胞和新生血管,改善心臟功能。
[Abstract]:[objective] to investigate the effect of basic fibroblast growth factor (bFGF) on the differentiation of adipose stem cells (ADSCs) into cardiomyocytes. [methods] ADSC was isolated, cultured and identified from SD rats. To evaluate the differentiation ability of ADSC, 60 rats were randomly divided into 4 groups: ADSC group, B FGF group, FGF group and b FGF ADSC group. After 1 week of myocardial infarction was established, PBSb FGFA ADSC and b FGF ADSCC were measured by echocardiography after 4 weeks of myocardial infarction. The left ventricular ejection fraction (LVEF) of rats with myocardial infarction was measured, and the infarct size of left ventricle was measured. Microvessel density (MVD) and ADSC differentiation. [results] ADSCs were successfully isolated. Flow cytometry showed that most ADSC expressed CD29 and CD90, but not CD34 and CD45.ADSC could be induced to adipose. The EF of osteoblast. B FGF ADSC group and ADSC group was higher than that of PBS group and b FGF group. The EF of FGF ADSC group was higher than that of ADSC group P0. 01 ADSC group and b FGF group. There was no significant difference in EF between b FGF ADSC group and b FGF group. The infarct area of b FGF ADSC group and ADSC group was smaller than that of PBS group and b FGF group. The infarct area of group B was smaller than that of group ADSC (P 0.01). There was no significant difference in infarct area between group B and group B FGF.
