【摘要】: Friedenstein等發(fā)現(xiàn)的骨髓MSCs有著巨大生物學(xué)潛能及應(yīng)用前景,而其高度污染性及隨年齡增加而擴(kuò)增與分化能力降低、創(chuàng)傷性操作等缺點(diǎn)限制了其廣泛使用。2003年HUC-MSCs的成功提取及相關(guān)研究證實(shí)其具有骨髓MSCs的特點(diǎn)并具有來源充分、提取簡(jiǎn)單、增殖旺盛、不悖倫理、無創(chuàng)操作等優(yōu)勢(shì)。但HUC-MSCs成為骨組織工程學(xué)種子細(xì)胞面臨是否存在同種異體移植排斥問題。全面研究其移植免疫學(xué)特性成為HUC-MSCs是否具有臨床應(yīng)用前景的關(guān)鍵。 本課題成功對(duì)HUC-MSCs進(jìn)行提取,觀察其生物學(xué)特征及體外成骨誘導(dǎo),對(duì)誘導(dǎo)體系的變化進(jìn)行動(dòng)態(tài)觀察,檢測(cè)其不同條件下對(duì)同種異體白細(xì)胞抗原、B7因子的表達(dá),設(shè)定多重因素下的MLR,對(duì)臨床應(yīng)用前后及MLR反應(yīng)前后T細(xì)胞亞型及細(xì)胞因子變化進(jìn)行分析,并對(duì)其免疫學(xué)機(jī)制進(jìn)行探討,為臨床應(yīng)用提供直接的理論依據(jù)。結(jié)論:HUC-MSCs為MSCs家族中一員并具有增殖旺盛、傳代穩(wěn)定、體外誘導(dǎo)成骨高效、同種異體移植的免疫逃逸及免疫調(diào)節(jié)能力,可能成為骨組織工程學(xué)理想的種子細(xì)胞。
[Abstract]:Bone marrow MSCs discovered by Friedenstein et al has great biological potential and application prospect, and its high pollution, amplification and differentiation ability decrease with age. In 2003, the successful extraction of HUC-MSCs and related studies confirmed that it has the characteristics of bone marrow MSCs and has sufficient source, simple extraction, vigorous proliferation, not contrary to ethics. However, HUC-MSCs is faced with the problem of allograft rejection or not. It is the key to study the immunological characteristics of HUC-MSCs transplantation for its clinical application prospect. In this study, HUC-MSCs was extracted successfully, its biological characteristics and osteogenic induction in vitro were observed, the changes of induction system were observed dynamically, and the expression of allogeneic leukocyte antigen factor B7 was detected under different conditions. The changes of T cell subtypes and cytokines before and after clinical application and MLR reaction were analyzed, and the immunological mechanism was discussed. Conclusion\\\. May be an ideal seed cell for bone tissue engineering.
