年輕骨髓間充質(zhì)干細胞可通過細胞融合改善年老骨髓間充質(zhì)干細胞功能

【摘要】:目的:誘導(dǎo)年輕與年老小鼠骨髓間充質(zhì)干細胞(BMSCs)融合,探討融合后年輕BMSCs對年老BM-SCs功能的影響。方法:年輕(2-3月齡)及年老(18-24月齡)C57BL/6小鼠BMSCs在紅色熒光染料PKH26標(biāo)記后分別與年輕及年老綠色熒光蛋白轉(zhuǎn)基因C57BL/6小鼠BMSCs通過聚乙烯二醇(PEG1500)誘導(dǎo),建立細胞融合模型。通過流式細胞儀鑒定細胞融合率和細胞表面特異性抗原的表達。免疫熒光顯微鏡觀察融合細胞形態(tài)及細胞核特性。通過細胞計數(shù)比較5組細胞(年輕組Y,年老組O,年輕融合組Y-Y,年輕年老融合組Y-O,年老融合組O-O)在2d、4d、6d、8d的細胞增殖能力。誘導(dǎo)細胞向成骨細胞及脂肪細胞分化,比較5組細胞的分化能力。結(jié)果:通過PEG誘導(dǎo),可獲得30.45%±4.13%的融合細胞,3組不同年齡融合組的細胞融合率無顯著差異。融和細胞表達間充質(zhì)干細胞表面特異性抗原CD44、Sca-1,而不表達造血干細胞表面特異性抗原CD34、CD117、CD31、CD45。在2d、4d、6d、8d,Y-O組細胞數(shù)量增加百分率比O-O組均顯著增高。代表成骨細胞分化能力強弱的茜素紅染色陽性率和代表脂肪細胞分化能力強弱的油紅O染色陽性率,Y-O組比O-O組均顯著增高,具體數(shù)值分別為:(25.46%±1.52%)vs(13.85%±1.69%),P0.01;(12.99%±2.61%)vs(6.03%±1.71%),P0.05。結(jié)論:年輕BMSCs可通過細胞融合改善年老BMSCs增殖及分化功能。
[Abstract]:Objective: to induce the fusion of bone marrow mesenchymal stem cells (BMSCs) in young and aged mice. To investigate the effect of young BMSCs on the function of aged BM-SCs after fusion. Methods: the BMSCs of young and aged C57BL / 6 mice were labeled with red fluorescent dye PKH26 and BMSCs of young and aged C57BL / 6 mice were treated with green fluorescent protein transgenic C57BL / 6 mice. Induced by PEG1500). Cell fusion model was established. Cell fusion rate and cell surface specific antigen expression were identified by flow cytometry. Morphology and nuclear characteristics of fusion cells were observed by immunofluorescence microscope. 5 groups of cells were compared by cell count. The proliferation ability of cells (young group, old group, young fusion group, young fusion group, young and old fusion group, young fusion group, old fusion group, O-O group) was induced to differentiate into osteoblasts and adipocytes at 2 d, 4 d, 6 d and 6 d, respectively. Results: the differentiation ability of 5 groups of cells was induced by PEG. There was no significant difference in cell fusion rate among the fusion cells of 30.45% 鹵4.13% and different age groups. The fusion cells expressed mesenchymal stem cell surface specific antigen CD44-Sca-1, but did not express hematopoietic stem cell surface specific antigen CD34, CD117, CD31 and CD45. The fusion cells expressed the surface specific antigen of mesenchymal stem cells for 6 days and 8 days after 2 days. The positive rate of alizarin red staining representing the differentiation ability of osteoblasts and the positive rate of oil red O staining representing the differentiation ability of adipocytes were significantly higher in Y-O group than in O-O group. The specific values were 25.46% 鹵1.52% + 13.85% 鹵1.69% 鹵1.69% and 12.99% 鹵2.61% 鹵6.03% 鹵1.71% respectively. Conclusion: young BMSCs can improve the proliferation and differentiation of aged BMSCs through cell fusion.
