電離輻射對小鼠免疫器官調(diào)節(jié)性T細(xì)胞及其相關(guān)分子表達的影響

【摘要】: 本文初步研究了調(diào)節(jié)性T細(xì)胞(Treg細(xì)胞)及其相關(guān)分子Foxp3和TGF-β1在不同時間和不同劑量電離輻射作用下,從基因水平、蛋白水平和細(xì)胞水平探討其表達的時間效應(yīng)和劑量效應(yīng)變化。實驗結(jié)果顯示,在胸腺細(xì)胞中CD4+CD25+細(xì)胞數(shù)不管在低劑量還是高劑量輻射作用下均增多,但脾細(xì)胞中的CD4+CD25+細(xì)胞數(shù)只在高劑量輻射作用下明顯增多;這很可能和脾細(xì)胞中含有的T淋巴細(xì)胞以成熟T細(xì)胞為主有關(guān)。 Foxp3是Treg細(xì)胞的特異性標(biāo)志,對Treg細(xì)胞的分化有著重要作用,尤其是對Treg細(xì)胞功能的發(fā)揮是必不可少的;本實驗提示,在低劑量照射條件下其表達沒有上調(diào),甚至略有下降,這也說明了在低劑量照射時Treg細(xì)胞的免疫負(fù)性調(diào)節(jié)功能沒有激活或上調(diào)。高劑量照射條件下Foxp3表達明顯上調(diào),提示Treg細(xì)胞被激活,行使免疫負(fù)性調(diào)節(jié)。因此,不同劑量電離輻射對機體的免疫抑制反應(yīng)的關(guān)鍵可能在Foxp3分子的變化上。 TGF-β1在CD4+CD25+Treg細(xì)胞的發(fā)育分化成熟中起到了決定性的作用,在本實驗中TGF-β1與CD4+CD25+Treg細(xì)胞不論是在時間效應(yīng)還是在劑量效應(yīng)變化中,都有類似的變化,這在一定程度上解釋了Treg細(xì)胞在輻射條件下表達變化的趨勢,這印證了TGF-β1對CD4+CD25+Treg細(xì)胞的發(fā)育成熟所起到的作用;大劑量輻射作用下CD4+CD25+TGF-β1促進Treg細(xì)胞的數(shù)量增加,為其抑制功能增強提供了條件。
[Abstract]:In this paper, we preliminarily studied regulatory T cell / Treg cell) and its related molecules Foxp3 and TGF- 尾 _ 1 at different time and different doses of ionizing radiation. The effects of time and dose on the expression of CD4 CD25 in thymocytes were investigated at the protein and cell levels. The results showed that the number of CD4 CD25 cells in thymocytes was increased in both low and high doses of radiation. However, the number of CD4 CD25 cells in spleen cells increased only under high dose radiation, which may be related to the majority of T lymphocytes contained in spleen cells. Foxp3 is a specific marker of Treg cells and plays an important role in the differentiation of Treg cells, especially the function of Treg cells. This also indicates that the immuno-negative regulatory function of Treg cells is not activated or upregulated under low dose irradiation. The expression of Foxp3 is obviously up-regulated under high dose irradiation, suggesting that Treg cells are activated and exercise immuno-negative regulation. The key to the immunosuppressive response of different doses of ionizing radiation may lie in the changes of Foxp3 molecules. TGF- 尾 1 plays a decisive role in the development, differentiation and maturation of CD4 CD25 Treg cells. In this experiment, TGF- 尾 1 and CD4 CD25 Treg cells have similar changes in both time and dose effects. This may explain to some extent the tendency of expression change of Treg cells under radiation, which confirms the effect of TGF- 尾 1 on the development and maturation of CD4 CD25 Treg cells, and CD4 CD25 TGF- 尾 1 promotes the increase of Treg cells under high dose radiation. It provides the condition for the enhancement of its inhibitory function.
