【摘要】：脾虛證是臨床常見(jiàn)的中醫(yī)證候之一,是慢性消化系統(tǒng)疾病的主要證型。本研究將ERIC-PCR和16S rDNA-RFLP兩種分子生物學(xué)方法聯(lián)合應(yīng)用,分析醋酸造模脾虛大鼠腸道菌群變化特征,從腸道菌群角度探索脾虛證本質(zhì)。 本研究通過(guò)醋酸灌胃塑造醋酸脾虛證大鼠模型,從大鼠糞便中直接提取腸道菌群總DNA,用ERIC及16S rDNA引物對(duì)其進(jìn)行擴(kuò)增,檢測(cè)腸道菌群的總體存在及變化情況,分別建立大鼠腸道菌群ERIC-PCR指紋圖譜及16S rDNA-RFLP酶切指紋圖譜;確定醋酸脾虛模型大鼠腸道菌群基因指紋特征及脾虛證治療藥物的評(píng)價(jià)指標(biāo),包括圖譜中特征條帶的凈面積(豐度)、多樣性指數(shù)和相似性指數(shù)。采用建立的分子指標(biāo)分別比較四君子湯、理中湯、補(bǔ)中益氣湯及益胃湯四種臨床常用脾虛治療中藥復(fù)方對(duì)模型大鼠的治療作用,建立了藥效評(píng)價(jià)分子指標(biāo)。 本研究將分子生物學(xué)和基因組學(xué)方法運(yùn)用于脾虛證及其相關(guān)治療中藥的評(píng)價(jià)與篩選中,并建立了相關(guān)的分子評(píng)價(jià)指標(biāo),對(duì)進(jìn)一步探討脾虛證的本質(zhì)及其治療中藥的藥效及篩選提供了新的思路與方法。
[Abstract]:Spleen deficiency is one of the common clinical syndromes, is the main syndrome of chronic diseases of the digestive system. The combined application of ERIC-PCR and 16S rDNA-RFLP two kinds of molecular biology methods, analysis of acetic acid model of spleen deficiency rats showed intestinal flora changes, explore the essence of spleen deficiency from the intestinal flora.
This study by intragastric administration of acetic acid acetic acid model of spleen deficiency rats, direct extraction of total DNA from intestinal flora in feces of rats, they were amplified by ERIC and 16S rDNA were detected the overall presence of the intestinal microflora and changes, respectively establish the rat intestinal flora ERIC-PCR fingerprint and 16S fingerprint rDNA-RFLP enzyme atlas; determine the index of spleen deficiency rat model of acetic acid bacteria gene fingerprint characteristics and spleen deficiency treatment, including the net area of characteristic bands (abundance), diversity index and similarity index. The molecular index is established to compare the Four Gentlemen Decoction and Lizhong decoction, Buzhongyiqi Decoction and Yi the stomach soup of four kinds of commonly used clinical therapeutic effect of Chinese medicine compound on spleen deficiency rat model, established the efficacy evaluation of molecular markers.
This study will be applied to screening and evaluation of spleen deficiency and related Chinese medicine treatment methods in molecular biology and genomics, and established the relevant molecular evaluation index, and further explore the essence of the efficacy of screening of spleen deficiency and the treatment of traditional Chinese medicine provides ideas and new methods.
