【摘要】: 視覺系統(tǒng)一些疾病會(huì)造成視神經(jīng)損傷而引起患者失明,目前臨床上尚無良好的治療方法,而視神經(jīng)再生的研究將為治療這些疾病提供實(shí)驗(yàn)依據(jù)。本研究通過體外培養(yǎng)妊娠晚期大鼠羊膜上皮細(xì)胞,予以免疫細(xì)胞化學(xué)、RT-PCR等方法鑒定后,應(yīng)用不含血清的神經(jīng)干細(xì)胞培養(yǎng)基誘導(dǎo)大鼠羊膜上皮細(xì)胞向類神經(jīng)干細(xì)胞方向分化,并予以免疫細(xì)胞化學(xué)、RT-PCR等方法鑒定。然后,將Hoechst33342熒光標(biāo)記的正常羊膜上皮細(xì)胞與誘導(dǎo)后的羊膜上皮細(xì)胞分別移植入大鼠受損視神經(jīng)中,應(yīng)用Hoechst33342熒光追蹤移植細(xì)胞、視網(wǎng)膜HE染色和尼氏染色計(jì)數(shù)視網(wǎng)膜節(jié)細(xì)胞數(shù)量、視神經(jīng)HE染色、免疫組織化學(xué)染色等方法初步探討細(xì)胞移植后對視網(wǎng)膜節(jié)細(xì)胞的存活和視神經(jīng)軸突再生所產(chǎn)生的作用。結(jié)果表明,應(yīng)用不含血清的神經(jīng)干細(xì)胞培養(yǎng)基可誘導(dǎo)大鼠羊膜上皮細(xì)胞向類神經(jīng)干細(xì)胞方向分化;誘導(dǎo)前后的羊膜上皮細(xì)胞移植入受損視神經(jīng)后,能在損傷部位大量存活并向損傷區(qū)兩端遷移,能提高視網(wǎng)膜節(jié)細(xì)胞存活率,增加遠(yuǎn)側(cè)段視神經(jīng)的細(xì)胞數(shù)量,促進(jìn)視神經(jīng)軸突再生。相對來說,誘導(dǎo)后的羊膜上皮細(xì)胞較誘導(dǎo)前的細(xì)胞移植對視神經(jīng)修復(fù)作用更強(qiáng)些。從而提示我們誘導(dǎo)后的羊膜上皮細(xì)胞經(jīng)移植后能促進(jìn)視神經(jīng)損傷的修復(fù),為臨床修復(fù)視神經(jīng)提供新思路。
[Abstract]:Some diseases of the visual system can cause blindness in patients with optic nerve injury, and there is no good treatment in clinic. The study of optic nerve regeneration will provide experimental basis for the treatment of these diseases. In this study, amniotic epithelial cells were cultured in vitro and identified by immunocytochemistry and RT-PCR. Rat amniotic epithelial cells were induced to differentiate into neural stem cells (NSCs) by using serum-free neural stem cells (NSCs) medium, and were identified by immunocytochemistry and RT-PCR. Hoechst33342 fluorescent labeled normal amniotic epithelial cells and induced amniotic epithelial cells were transplanted into the injured optic nerve of rats. The number of retinal ganglion cells and optic nerve HE staining were counted by Hoechst33342 fluorescence tracing, HE staining and Nissl staining of retina. The effects of cell transplantation on the survival of retinal ganglion cells and the regeneration of optic nerve axons were preliminarily investigated by immunohistochemical staining. The differentiation of rat amniotic epithelial cells into neural stem cells was induced by using neural stem cell culture medium without serum. After transplantation of amniotic epithelial cells into the injured optic nerve before and after induction, they can survive and migrate to both ends of the injured area, which can improve the survival rate of retinal ganglion cells and increase the number of distal optic nerve cells. Promote axonal regeneration of the optic nerve. Relatively speaking. The effect of induced amniotic epithelial cells on optic nerve repair is stronger than that of pre-induced amniotic epithelial cells, which suggests that the induced amniotic epithelial cells can promote the repair of optic nerve injury after transplantation.
