【摘要】： 目的:鑒定抗云錫礦工肺鱗癌單克隆抗體(C6)的特異性、純化其抗體,為進(jìn)一步獲取YTMLC-90(云錫礦工肺鱗癌)腫瘤抗原蛋白及腫瘤抗原基因、探索其發(fā)生機(jī)制奠定基礎(chǔ)。 方法:采用免疫印跡(Western blot),檢測各種腫瘤細(xì)胞、人正常細(xì)胞和各種腫瘤組織及正常組織與抗云錫礦工肺鱗癌單克隆抗體(C6)的交叉反應(yīng)。采用親和層析法純化該抗體;用免疫印跡方法鑒定純化單抗。 結(jié)果:抗云錫礦工肺鱗癌單克隆抗體(C6)與各種肺癌細(xì)胞及肺癌組織抗原有交叉反應(yīng),而與其它腫瘤細(xì)胞、腫瘤組織及人正常細(xì)胞、正常組織抗原無交叉反應(yīng)。純化出了抗云錫礦工肺鱗癌單抗(C6)。 結(jié)論:抗云錫礦工肺鱗癌單克隆抗體(C6)與肺癌相關(guān)抗原有較高的特異性,而與其它腫瘤相關(guān)抗原及正常組織抗原無特異性。獲得有活性的純化單抗。
[Abstract]:Objective: to identify the specificity of monoclonal antibody (C6) against lung squamous cell carcinoma (LSCC) of Yunnan tin miners and to purify its antibody in order to obtain tumor antigen protein and tumor antigen gene of YTMLC-90 (Yunxi miner lung squamous cell carcinoma). It lays a foundation for exploring the mechanism of its occurrence. Methods: Western blotters were used to detect various tumor cells. The cross-reaction of human normal cells, various tumor tissues and normal tissues with monoclonal antibody (C6) against Yunnan tin miners' lung squamous cell carcinoma was used to purify the antibody by affinity chromatography. The purified McAb was identified by Western blot. Results: the monoclonal antibody against lung squamous cell carcinoma (C6) was cross-reacted with various lung cancer cells and lung cancer tissue antigens, but with other tumor cells, tumor tissues and human normal cells. There was no cross reaction of normal tissue antigen. The monoclonal antibody against lung squamous cell carcinoma of Yuntin miners was purified. Conclusion: the monoclonal antibody (C6) against Yuntin miners' lung squamous cell carcinoma has high specificity with lung cancer associated antigens, but has no specificity with other tumor associated antigens and normal tissue antigens.
