【摘要】:目的:本課題以正常成年豚鼠膀胱Cajal樣間質(zhì)細(xì)胞為研究對(duì)象,通過對(duì)膀胱Cajal樣間質(zhì)細(xì)胞(interstitial cells of Cajal, ICCs)超微結(jié)構(gòu)觀察及電生理學(xué)特性研究,探討其是否為膀胱的起博細(xì)胞,為臨床上治療某些疾病(如detrusor instability, DI)提供新的切入點(diǎn)。 方法:本實(shí)驗(yàn)運(yùn)用采用酶消化法,得到體外培養(yǎng)狀態(tài)下的ICCs樣細(xì)胞,透射電鏡觀察正常成年豚鼠膀胱ICCs樣細(xì)胞的超微結(jié)構(gòu);采用激光共聚焦顯微鏡成像技術(shù)記錄Fluo4AM負(fù)載后的Cajal樣間質(zhì)細(xì)胞內(nèi)鈣離子熒光強(qiáng)度的變化,以熒光強(qiáng)度代表細(xì)胞內(nèi)鈣離子濃度的變化。;運(yùn)用膜片鉗技術(shù),在全細(xì)胞記錄模式下觀察其是否具有鈣電流,以驗(yàn)證其是否為膀胱的起博細(xì)胞。 結(jié)果:透射電鏡觀察到CCs樣細(xì)胞胞漿內(nèi)線粒體、粗面內(nèi)質(zhì)網(wǎng)較豐富,滑面內(nèi)質(zhì)網(wǎng)分散分布,中間絲難能見到,未見微管;運(yùn)用激光共聚焦顯微鏡成像技術(shù)連續(xù)掃描,見體外培養(yǎng)的原代膀胱Cajal樣間質(zhì)細(xì)胞有2種類型鈣震蕩:一類振幅大、頻率慢而規(guī)則的鈣震蕩;另一類振幅小、頻率快且不規(guī)則,且形成網(wǎng)絡(luò)的膀胱Cajal樣間質(zhì)細(xì)胞的鈣離子震蕩的幅度明顯高于單個(gè)的膀胱Cajal樣間質(zhì)細(xì)胞。膀胱逼尿肌細(xì)胞鈣震蕩振幅小不規(guī)則,其振幅顯著低于Cajal樣間質(zhì)細(xì)胞第一類鈣震蕩振幅;膜片鉗試驗(yàn)膀胱Cajal樣細(xì)胞的鈣離子電流不明顯。 結(jié)論:電鏡觀察到Cajal樣細(xì)胞具有獨(dú)特的超微結(jié)構(gòu),與胃腸道中ICCs的超微結(jié)構(gòu)有相似;部分膀胱Cajal樣細(xì)胞具有自發(fā)而規(guī)律的鈣震蕩,類似消化道起搏細(xì)胞Cajal間質(zhì)細(xì)胞(interstitial cells of Cajal,ICCs)的自發(fā)性鈣震蕩,膀胱逼尿肌無自發(fā)性鈣震蕩。提示:部分膀胱Cajal樣細(xì)胞是膀胱收縮活動(dòng)的起搏細(xì)胞;另外部分膀胱Cajal樣細(xì)胞可能起到傳導(dǎo)、耦合信息的作用。
[Abstract]:Objective: to study the Cajal like stromal cells of normal adult guinea pig bladder. The ultrastructure and electrophysiological characteristics of interstitial cells of Cajal (ICCs) were observed. To investigate whether it is a primordial cell of the bladder and to provide a new entry point for the clinical treatment of certain diseases such as detrusor instability. Methods: ICCs like cells were obtained by enzyme digestion in vitro. The ultrastructure of ICCs like cells in normal adult guinea pig bladder was observed by transmission electron microscope. Laser confocal microscopy (LSCM) was used to record the changes of intracellular calcium fluorescence intensity in Cajal like stromal cells after Fluo4AM loading, and the fluorescence intensity represented the change of intracellular calcium concentration. The patch-clamp technique was used to observe whether it had calcium current in whole cell recording mode to verify whether it was a pacemaker cell of the bladder. Results: the mitochondria in the cytoplasm of CCs like cells were observed by transmission electron microscope, the rough endoplasmic reticulum was abundant, the smooth endoplasmic reticulum was scattered, the middle filaments could not be seen, and no microtubules were found. Continuous scanning with laser confocal microscopy showed that there were two types of calcium oscillations in primary bladder Cajal like stromal cells: one was large amplitude, slow frequency and regular calcium oscillation; The other has small amplitude, fast frequency and irregular frequency. The amplitude of calcium oscillation in the mesenchymal cells of bladder Cajal was significantly higher than that in the single interstitial cells of Cajal. The amplitude of calcium oscillation in the bladder detrusor muscle cells was small and irregular. The amplitude of calcium oscillation was significantly lower than that of Cajal like stromal cells. Patch clamp test showed that calcium current in Cajal-like cells of bladder was not obvious. Conclusion: the ultrastructure of Cajal like cells is similar to that of ICCs in gastrointestinal tract. Some Cajal like cells of bladder have spontaneous and regular calcium concussion. Spontaneous calcium concussion similar to interstitial cells of Cajal interstitial cells in digestive tract pacemaker cells. There was no spontaneous calcium concussion in the detrusor of the bladder. It was suggested that some of the bladder Cajal like cells were pacemaker cells of bladder contraction. Another part of bladder Cajal-like cells may play a role in the transmission and coupling of information.
