【摘要】：目的研究龍血素A對(duì)白色假絲酵母菌生物膜的抑菌作用,并初步研究龍血素A是否通過(guò)影響或干擾白色假絲酵母菌毒力因子的轉(zhuǎn)錄和表達(dá)實(shí)現(xiàn)的。方法建立白色假絲酵母菌生物膜體外模型,MTT法計(jì)算龍血素A對(duì)白色假絲酵母菌生物膜的抑制率;激光共聚焦觀察龍血素A對(duì)不同時(shí)間白色假絲酵母菌生物膜的抑菌效果;RT-PCR技術(shù)檢測(cè)藥物作用下白色假絲酵母菌毒力因子表達(dá)水平的變化。結(jié)果隨著藥物濃度的增加,龍血素A對(duì)白色假絲酵母菌生物膜的抑菌作用增強(qiáng)。激光共聚焦觀察顯示龍血素A使白色假絲酵母菌生物膜內(nèi)活菌死菌比例下降。RT-PCR結(jié)果表明龍血素A抑制了毒力因ALS3、SAP4的表達(dá)。結(jié)論龍血素A對(duì)白色假絲酵母菌生物膜有抑制作用,并在毒力因子ALS3、SAP4的表達(dá)過(guò)程中起到明顯的抑制作用。
[Abstract]:Objective to study the bacteriostatic effect of dragon blood A on Candida albicans biofilm. And the preliminary study on whether dragon blood A by affecting or interfering with the virulence factor transcription and expression of Candida albicans. Methods the model of Candida albicans biofilm in vitro was established. The inhibitory rate of dragonin A on Candida albicans biofilm was calculated by MTT method. Laser confocal focusing was used to observe the bacteriostatic effect of dragon blood A on the biofilm of Candida albicans at different time. The expression of virulence factor in Candida albicans was detected by RT-PCR. The bacteriostatic effect of dragon blood A on the biofilm of Candida albicans was enhanced. Laser confocal observation showed that the proportion of live bacteria in the biofilm of Candida albicans was decreased by dragon blood A. RT-PCR results showed that dragon blood in the biofilm of Candida albicans was reduced. A inhibited virulence factor ALS3. Conclusion Dragonin A can inhibit the biofilm of Candida albicans and play a significant role in the expression of virulence factor ALS3 / SAP4.
