【摘要】:目的觀察雙氫睪酮對(duì)原代培養(yǎng)大鼠Sertoli細(xì)胞GDNF及相關(guān)細(xì)胞信號(hào)通路關(guān)鍵蛋白的表達(dá),探討Sertoli細(xì)胞GDNF表達(dá)調(diào)控的機(jī)制。方法原代培養(yǎng)出生后20 d大鼠Sertoli細(xì)胞,給予雙氫睪酮干預(yù),免疫熒光檢測(cè)GDNF在細(xì)胞中定位和表達(dá)強(qiáng)度,Western blot檢測(cè)GDNF,ERK和CREB蛋白的表達(dá)及磷酸化情況,并用ERK和c AMP/PKA信號(hào)通路抑制劑干預(yù)。結(jié)果 GDNF表達(dá)在Sertoli細(xì)胞質(zhì)中,雙氫睪酮干預(yù)可增加GDNF的表達(dá)(P0.05),并使ERK和CREB磷酸化水平增高(P0.05),使用ERK和c AMP/PKA信號(hào)通路抑制劑可減低雙氫睪酮誘導(dǎo)的GDNF表達(dá)(P0.05)。結(jié)論雙氫睪酮通過(guò)激活ERK信號(hào)通路誘導(dǎo)Sertoli細(xì)胞GDNF的表達(dá)。
[Abstract]:Objective to investigate the effect of dihydrotestosterone on the expression of GDNF and related cell signal pathway key proteins in primary cultured rat Sertoli cells. To explore the mechanism of GDNF expression and regulation in Sertoli cells. Methods Sertoli cells were cultured 20 days after birth and treated with dihydrotestosterone. The localization and expression intensity of GDNF in the cells were detected by immunofluorescence. The expression and phosphorylation of GDNF ERK and CREB protein were detected by Western blot. ERK and c AMP/PKA signaling pathway inhibitors were used to intervene. Results GDNF was expressed in the cytoplasm of Sertoli. Dihydrotestosterone could increase the expression of GDNF and increase the phosphorylation level of ERK and CREB (P0.05). Using ERK and c AMP/PKA signaling pathway inhibitors can reduce GDNF expression induced by dihydrotestosterone (DHT). Conclusion dihydrotestosterone induces GDNF expression in Sertoli cells by activating ERK signaling pathway.
