【摘要】：【目的】 研究大鼠脂肪干細(xì)胞(ADSCs)體外單層培養(yǎng)條件下誘導(dǎo)分化為平滑肌樣細(xì)胞的可行性。 【方法】 從SD大鼠的腹股溝脂肪墊分離獲取脂肪干細(xì)胞,測(cè)定其生長(zhǎng)曲線(xiàn)。取第4代細(xì)胞用成脂誘導(dǎo)液誘導(dǎo)分化,并用油紅O染色鑒定。取第4代細(xì)胞用成骨誘導(dǎo)液誘導(dǎo)分化,并用Von Kossa染色鑒定。取第4代細(xì)胞用含有β-巰基乙醇的成平滑肌誘導(dǎo)液誘導(dǎo),并用免疫組化的方法檢測(cè)α平滑肌肌動(dòng)蛋白(α-SMA)的表達(dá)。 【結(jié)果】 脂肪干細(xì)胞成梭形和多角形,體外生長(zhǎng)迅速,生長(zhǎng)曲線(xiàn)表明傳代2天后細(xì)胞進(jìn)入對(duì)數(shù)生長(zhǎng)期。第4代脂肪干細(xì)胞,成脂誘導(dǎo)后,油紅O染色證實(shí)細(xì)胞內(nèi)存在脂滴;成骨誘導(dǎo)后,Von Kossa染色證實(shí)有鈣化結(jié)節(jié)。脂肪干細(xì)胞誘導(dǎo)平滑肌樣細(xì)胞免疫組化結(jié)果,含β-巰基乙醇誘導(dǎo)液誘導(dǎo)組和未誘導(dǎo)組細(xì)胞α-SMA的表達(dá)陽(yáng)性率分別為(29.80±6.89)%、(2.89±1.24)%。誘導(dǎo)組細(xì)胞α-SMA的表達(dá)陽(yáng)性率高于未誘導(dǎo)組,差異有統(tǒng)計(jì)學(xué)意義(P0.01)。 【結(jié)論】 脂肪干細(xì)胞經(jīng)誘導(dǎo)后出現(xiàn)明顯的平滑肌細(xì)胞特征,可成為平滑肌相關(guān)疾病在組織工程研究上新的種子細(xì)胞來(lái)源。
[Abstract]:[purpose] To study the feasibility of differentiation into smooth muscle like cells (SMC) induced by rat adipose stem cells (ADSCs) in vitro monolayer culture. [methods] The adipose stem cells were isolated from the groin fat pad of SD rats and their growth curves were measured. The differentiation of the cells was induced by osteoblast inducer and Von Kossa staining. The fourth passage cells were induced by 尾 -mercaptoethanol. The expression of 偽-smooth muscle actin (偽 -SMA) was detected by immunohistochemistry. [results] Adipose stem cells were spindle-shaped and polygonal, and grew rapidly in vitro. The growth curve showed that the cells entered logarithmic growth phase 2 days after passage, and the fourth generation of adipose stem cells was induced by adipogenesis. Oil red O staining confirmed the presence of lipid droplets in the cells. Calcified nodules were confirmed by Von Kossa staining after osteogenic induction. Immunohistochemical results of smooth muscle like cells induced by adipose stem cells were observed. The positive rate of 偽 -SMA expression in cells induced by 尾 -mercaptoethanol was 29.80 鹵6.89%, respectively. The positive rate of 偽 -SMA expression in the induction group was higher than that in the uninduced group, and the difference was statistically significant (P 0.01). [conclusion] After induction, adipose stem cells have obvious characteristics of smooth muscle cells, which may be a new source of seed cells for tissue engineering of smooth muscle related diseases.
