豐富環(huán)境對缺血缺氧腦損傷新生大鼠海馬腦源性神經(jīng)營養(yǎng)因子表達及細胞凋亡的影響

【摘要】:目的:探討豐富環(huán)境對缺血缺氧腦損傷(HIBD)新生大鼠海馬腦源性神經(jīng)營養(yǎng)因子(BDNF)表達及細胞凋亡的影響。方法:采用隨機數(shù)字表法將36只7日齡新生大鼠分為假手術組、模型組及豐富環(huán)境組,各組又分為術后7d、28d兩個亞組(n=6)。采用Rice-Vannucci經(jīng)典方法建立新生大鼠HIBD模型。模型組及假手術組不予任何干預,豐富環(huán)境組予以早期撫觸及豐富環(huán)境刺激治療。術后7d、28d采用水迷宮檢測方法評估大鼠學習記憶能力,免疫組化法檢測大鼠海馬CA1區(qū)BDNF的表達,TUNEL法檢測海馬CA1區(qū)細胞凋亡。結(jié)果:術后7d、28d,與模型組比較,豐富環(huán)境組逃避潛伏期均縮短(P0.05),穿越平臺次數(shù)增多(P0.05),BDNF表達升高(P0.05),TUNEL陽性細胞數(shù)降低(P0.05)。結(jié)論:豐富環(huán)境能改善HIBD新生大鼠學習記憶能力,其機制可能與上調(diào)海馬區(qū)BDNF的表達及抑制細胞凋亡有關。
