人腦膠質(zhì)瘤中CD133、SSEA-1和Nestin的表達(dá)和意義

【摘要】:目的:膠質(zhì)瘤是腦腫瘤中最常見的腫瘤,約占50%。盡管目前的外科手術(shù),,放、化療都有了極大的提高,但是對于膠質(zhì)瘤的治療及預(yù)后,還遠(yuǎn)未達(dá)到到理想的效果。膠質(zhì)瘤干細(xì)胞的提出,給我們提供了一個(gè)全新的思路。本實(shí)驗(yàn)欲研究膠質(zhì)瘤干細(xì)胞標(biāo)志物CD133、SSEA-1(階段特異性胚胎抗原-1)和Nestin(神經(jīng)巢蛋白)在人腦膠質(zhì)瘤組織中表達(dá)及與病理分級的相關(guān)性;探討三者在人腦膠質(zhì)瘤的診斷及惡性程度判斷中的臨床意義,以及為治療提供分子生物學(xué)理論基礎(chǔ)。 方法:收集廣西醫(yī)科大學(xué)一附院(2011年9月—2012年12月)經(jīng)病理證實(shí)為膠質(zhì)瘤,按2007年WHO分級標(biāo)準(zhǔn):Ⅰ級6例,Ⅱ級16例,Ⅲ級13例,Ⅳ級19例,另外用6例正常腦組織為對照。應(yīng)用免疫組織化學(xué)的方法檢測54例腦膠質(zhì)瘤組織和6例正常腦組織標(biāo)本中CD133、SSEA-1及Nestin的表達(dá)。 結(jié)果:①54例腦膠質(zhì)瘤組織和6例正常腦組織標(biāo)本中CD133陽性細(xì)胞率率分別為25.38%和0%(P0.05),SSEA-1陽性細(xì)胞率分別為26.62%和0%,Nestin陽性細(xì)胞率分別為22.39%和0%。②對54例腦膠質(zhì)瘤組織中CD133和SSEA-1及Nestin陽性細(xì)胞率分別進(jìn)行多個(gè)病理級別之間的均數(shù)比較: I級和Ⅱ級之間差別無統(tǒng)計(jì)學(xué)意義(P>0.05),Ⅰ級和Ⅲ、Ⅳ級間差別具有統(tǒng)計(jì)學(xué)意義(P<0.01),Ⅱ級和Ⅲ、Ⅳ級間差別具有統(tǒng)計(jì)學(xué)意義(P<0.05), Ⅲ級和Ⅳ級間差別具有統(tǒng)計(jì)學(xué)意義(P<0.05)。③腦膠質(zhì)瘤組織中SSEA-1陽性細(xì)胞率大于CD133大于Nestin,差別具有統(tǒng)計(jì)學(xué)意義(P<0.05);SSEA-1陽性細(xì)胞率和CD133陽性細(xì)胞率呈正相關(guān)關(guān)系(Cp=0.84, P<0.05);CD133陽性細(xì)胞率和Nestin陽性細(xì)胞率呈正相關(guān)關(guān)系(Cp=0.48,P<0.05);SSEA-1陽性細(xì)胞率和Nestin陽性細(xì)胞率呈正相關(guān)系(Cp=0.44,P<0.05)。 結(jié)論:①人腦膠質(zhì)瘤組織中在不同病理級別的膠質(zhì)瘤標(biāo)本組中CD133、SSEA-1及Nestin表達(dá)程度不同; CD133、SSEA-1、Nestin的表達(dá)與膠質(zhì)瘤病理分級呈正相關(guān),提示三者的高表達(dá)與膠質(zhì)瘤的惡性程度有關(guān)。②檢測腦膠質(zhì)瘤組織中CD133、SSEA-1、Nestin的表達(dá)有助于膠質(zhì)瘤患者的診斷及惡性程度判斷。③CD133、SSEA-1和Nestin作為膠質(zhì)瘤干細(xì)胞標(biāo)記物在膠質(zhì)瘤的診斷和治療以及評估預(yù)后和未來個(gè)性化綜合治療中發(fā)揮重大的作用。

Although the current surgery, radiotherapy and chemotherapy have been greatly improved, but for the treatment and prognosis of glioma, it is far from the ideal effect. The proposal of glioma stem cells provides us with a new way of thinking. The purpose of this study was to study the expression of glioma stem cell markers CD133,SSEA-1 (stage specific embryonic antigen-1) and Nestin (nestin) in human glioma tissues and their correlation with pathological grade, to explore the clinical significance of the three markers in the diagnosis and malignant degree of human glioma, and to provide the theoretical basis of molecular biology for treatment.
