miRNA-210在新生大鼠缺氧缺血性腦水腫中的作用

【摘要】:目的探討mi RNA-210在新生大鼠缺氧缺血性腦水腫中的作用。方法將80只新生大鼠隨機(jī)分為對(duì)照組、生理鹽水組、mi RNA-210表達(dá)抑制組及mi RNA-210過表達(dá)組,每組20只;各組大鼠再隨機(jī)分為假手術(shù)組及缺氧缺血(HI)組,每組10只。HI組新生大鼠結(jié)扎左側(cè)頸總動(dòng)脈后,置于8%O2和92%N2的混合氣體缺氧艙中持續(xù)2 h;假手術(shù)組僅游離左側(cè)頸總動(dòng)脈,但不予結(jié)扎和缺氧處理。HI或假手術(shù)術(shù)后,分別向生理鹽水組、mi RNA-210表達(dá)抑制組和mi RNA-210過表達(dá)組大鼠顱內(nèi)注射生理鹽水(2.5 mg/kg),mi RNA-210incubitor(2.5 mg/kg)和mi RNA-210 minic(2.5 mg/kg),對(duì)照組大鼠不予任何處理。3 d后斷頭,取左側(cè)腦組織,采用熒光定量PCR方法檢測(cè)mi RNA-210的表達(dá);采用干濕質(zhì)量法檢測(cè)腦組織含水量;蘇木精-伊紅染色觀察腦組織形態(tài)學(xué)改變。結(jié)果各組HI新生大鼠腦組織中mi RNA-210的表達(dá)較相應(yīng)假手術(shù)組均顯著下調(diào),腦組織含水量顯著上調(diào)(P0.05)。與生理鹽水HI組比較,mi RNA-210表達(dá)抑制HI組中mi RNA-210的表達(dá)顯著下調(diào),腦組織含水量顯著上調(diào)(P0.05);mi RNA-210過表達(dá)HI組中mi RNA-210的表達(dá)顯著上調(diào),腦組織含水量顯著下調(diào)(P0.05)。蘇木精-伊紅染色結(jié)果提示mi RNA-210表達(dá)抑制HI組水腫改變明顯;mi RNA-210過表達(dá)HI組水腫改變明顯改善。結(jié)論新生大鼠腦HI后mi RNA-210表達(dá)下調(diào),可能參與了新生大鼠缺氧缺血性腦水腫的發(fā)生發(fā)展。
[Abstract]:Objective to investigate the role of mi RNA-210 in anoxic ischemic brain edema in neonatal rats. Methods 80 neonatal rats were randomly divided into control group, saline group, mi RNA-210 expression inhibition group and mi RNA-210 overexpression group with 20 rats in each group, and the rats in each group were randomly divided into pseudo-operation group (n = 10) and hypoxia-ischemic (HI) group (n = 10). In HI group, the left common carotid artery was ligated and placed in the mixed gas hypoxia chamber of 8%O2 and 92%N2 for 2 hours. The left common carotid artery was only free in the pseudo-operation group, but not treated with ligation and hypoxia. Hi or pseudo-operation, the rats in the saline group, the mi RNA-210 expression inhibition group and the mi RNA-210 overexpression group were injected with normal saline (2.5 mg/kg), mi RNA-210incubitor (2.5 mg/kg) and mi RNA-210 minic (2.5 mg/kg) respectively), while the rats in the control group were not treated with any treatment. 3 days later, the left brain tissue was decapitated and the left brain tissue was taken. The expression of mi RNA-210 was detected by fluorescence quantitative PCR. The water content of brain tissue was measured by dry and wet mass method, and the morphological changes of brain tissue were observed by hematoxylin and eosin staining. Results the expression of mi RNA-210 in brain tissue of HI neonatal rats in each group was significantly lower than that in the corresponding pseudo-operation group, and the water content of brain tissue was significantly up-regulated (P 0.05). Compared with saline HI group, the expression of mi RNA-210 in HI group was significantly down-regulated, and the water content of brain tissue was significantly up-regulated (P 0.05). The expression of mi RNA-210 in HI group was significantly up-regulated and the water content of brain tissue was significantly down-regulated (P 0.05). The results of hematoxylin and eosin staining suggested that the edema changes in HI group were significantly decreased by mi RNA-210 expression, and the edema changes in mi RNA-210 overexpression HI group were significantly improved. Conclusion the down-regulation of mi RNA-210 expression in neonatal rat brain after HI may be involved in the occurrence and development of hypoxic-ischemic brain edema in neonatal rats.
