Notch信號通路在大鼠骨髓間充質(zhì)干細胞移植后促進腦缺血區(qū)血管新生過程中的作用

【摘要】:目的探討Notch1信號通路在骨髓間充質(zhì)干細胞(MSCs)移植后促進腦缺血區(qū)血管新生過程中的作用。方法分離、培養(yǎng)SD大鼠MSCs,制作大鼠大腦中動脈栓塞(MCAO)模型,將試驗分為正常組、腦缺血后MSCs移植組、腦缺血后MSCs移植+rh NF-κB組、腦缺血對照組。腦組織冰凍切片行Ⅷ因子免疫組織熒光染色檢測各試驗組MSCs移植1、4、7、14 d后缺血皮質(zhì)區(qū)微血管數(shù)量。Western印跡檢測各試驗組MSCs移植1、4、7、14 d后缺血皮質(zhì)區(qū)血管內(nèi)皮生長因子(VEGF)165、Notch1、Hes1蛋白表達。結(jié)果缺血對照組、MSCs移植組和MSCs移植+rh NF-κB組在各時間點微血管數(shù)量及VEGF165、Notch1、Hes1蛋白表達均顯著高于正常組,從第14天起開始下降,3組的微血管數(shù)量及VEGF165、Notch1、Hes1蛋白表達依次上升,差異顯著(P0.01)。結(jié)論 MSCs移植可促進缺血區(qū)新生血管的形成,并可通過激活Notch信號通路促進缺血皮質(zhì)區(qū)VEGF165的表達,從而促進缺血區(qū)的血管新生。
