【摘要】：目的：通過檢測腦膜瘤組織中促紅細(xì)胞生成素肝細(xì)胞(erythropoietin-producing hepatocellular Eph)受體酪氨酸激酶A2的表達(dá)情況，從而探索其與腦膜瘤發(fā)生、分級(jí)及瘤周水腫的關(guān)系。方法：①腦膜瘤標(biāo)本共60例，均取自瀘州醫(yī)學(xué)院附屬醫(yī)院神經(jīng)外科2012年1月至2013年1月手術(shù)切除病理標(biāo)本。所選病例均為原發(fā)首次手術(shù)病例，術(shù)前未行藥物及放射治療，各腦膜瘤標(biāo)本的病理資料和臨床資料（包括影像資料）均完整。對照組25例，為內(nèi)減壓術(shù)后所取正常蛛網(wǎng)膜組織。②腦膜瘤病理標(biāo)本按世界衛(wèi)生組織（WHO）2007年標(biāo)準(zhǔn)對其進(jìn)行病理分級(jí)并分為兩組。③所有的對照組標(biāo)本均在分離人體后12小時(shí)之內(nèi)進(jìn)行冷凍、固定處理。④對腦膜瘤及正常腦膜組織進(jìn)行免疫組化鏈霉菌抗生物素蛋白-過氧化物酶連結(jié)（streptavidin-perosidase S-P）法染色處理，檢測其中的EphA2表達(dá)情況。⑤觀察并記錄結(jié)果。結(jié)果：①正常腦膜中，EphA2表達(dá)陰性17例，陽性8例；在腦膜瘤組織中陰性12例，陽性48例，兩者之間差異有顯著性（2=18.089，P=0.0000.05)。②Ⅰ級(jí)腦膜瘤組織中EphA2陰性10例，陽性41例；在Ⅱ級(jí)腦膜瘤組織中，EphA2陰性2例，陽性7例，Ⅰ級(jí)和Ⅱ級(jí)腦膜瘤組織中EphA2陽性表達(dá)率差異無顯著性（2=0.033，P=0.58＞0.05）。③Ⅰ級(jí)腦膜瘤伴有瘤周水腫的有21例，水腫率41.2%，Ⅱ級(jí)腦膜瘤伴有瘤周水腫的有8例，水腫率88.9%，差異有顯著性（2=6.974，P=0.010.05）。④在腦膜瘤中，EphA2陽性的瘤周水腫率與陰性的瘤周水腫率差異無顯著性（2=1.352，P=0.201＞0.05）。結(jié)論：①腦膜瘤組織中EphA2呈過度表達(dá)，，提示EphA2可能參與了腦膜瘤的發(fā)生發(fā)展。②本實(shí)驗(yàn)結(jié)果提示EphA2與腦膜瘤的分級(jí)無關(guān)，原因可能與實(shí)驗(yàn)中Ⅱ級(jí)腦膜瘤總例數(shù)少有關(guān)，有待我們做進(jìn)一步的研究。③Ⅱ級(jí)腦膜瘤較Ⅰ級(jí)腦膜瘤更易發(fā)生瘤周水腫。④本實(shí)驗(yàn)結(jié)果提示EphA2與腦膜瘤瘤周水腫無關(guān)，可能同樣與實(shí)驗(yàn)中Ⅱ級(jí)腦膜瘤總例數(shù)少有關(guān)。
[Abstract]:Aim: to investigate the expression of erythropoietin (erythropoietin-producing hepatocellular Eph) receptor tyrosine kinase A2 in meningiomas and its relationship with meningioma occurrence, grading and peritumoral edema. Methods: 1 A total of 60 meningioma specimens were obtained from neurosurgery department of affiliated Hospital of Luzhou Medical College from January 2012 to January 2013. All the selected cases were primary operation cases. No drug and radiotherapy were performed before operation. The pathological and clinical data (including imaging data) of all meningioma specimens were complete. Control group 25 cases, Normal arachnoid tissue was taken after internal decompression. 2 pathological specimens of meningioma were classified according to WHO (WHO) 2007 standard and divided into two groups. Frozen within hours, 4 Immunohistochemical staining of streptomycin and peroxidase (streptavidin-perosidase S-P) was used to detect the expression of EphA2 in meningiomas and normal meningeal tissues. 5 the results were observed and recorded.
