牛磺酸對匹羅卡品致癇大鼠海馬苔蘚纖維出芽的影響

【摘要】:目的:研究;撬釋ζチ_卡品致癇大鼠海馬苔蘚纖維出芽的影響。 方法:1、制作匹羅卡品癲癇動物模型。將致癇鼠隨機(jī)分為三組,空白組、;撬嶂委熃M及治療對照組,每組10只。2、牛磺酸治療組于6天每天腹腔注射2g/kg;撬。觀察;撬嶂委熃M于不同時(shí)間點(diǎn)的癇性發(fā)作行為學(xué)改變,比較其癇性發(fā)作持續(xù)時(shí)間及發(fā)作分級的差異。行為學(xué)觀察參照Racine發(fā)作分級標(biāo)準(zhǔn),制訂行為學(xué)觀察表,觀察造模后各組大鼠癇性發(fā)作行為表現(xiàn),以造模后至首次出現(xiàn)Racine標(biāo)準(zhǔn)Ⅲ級以上發(fā)作時(shí)間計(jì)算潛伏期,分別記錄大鼠面部陣攣、節(jié)律點(diǎn)頭/甩尾、前肢陣攣、后肢站立/全身抽動、竄動/跌倒的次數(shù)。Ⅳ級以上大發(fā)作1h后腹腔注射10%水合氯醛(300mg/kg)終止發(fā)作,若注射水合氯醛30min后沒有終止發(fā)作,予150mg/kg劑量重復(fù)注射至終止(以3次為最高限)。3、行為學(xué)觀察結(jié)束后,觀察致癇鼠海馬組織中苔蘚纖維出芽情況。Nissl染色:切片經(jīng)二甲苯、梯度酒精脫脂至水,去離子水沖洗3min,1%甲苯胺藍(lán)60℃C浸染30min,70%乙醇2min,95%乙醇分化(鏡下控制,以尼氏小體顯示清晰為度),無水乙醇快速脫水,二甲苯透明,中性樹膠封片。Timm染色:運(yùn)用改良的Timm方法,切片依次浸入0.1%Na2S(溶于0.01mo1/L PBS),3%戊二醛各1h,然后再回到0.1%Na2S溶液浸泡1h。在暗室內(nèi)將晾干的切片入新鮮配置Timm顯色液中孵育30mmin,(顯色液的配置:50%阿拉伯膠粉60mL,檸檬酸緩沖液10mL(pH=3),5.6%對苯二酚30mL,17%硝酸銀1mL,硝酸銀最后加入)。暗室中去離子水沖洗5min,再流水沖洗20min,蘇木素復(fù)染15-30s,常規(guī)梯度脫水透明封片。實(shí)驗(yàn)數(shù)據(jù)采用SPSS17.0軟件進(jìn)行統(tǒng)計(jì)處理,結(jié)果以x±s表示,采用單因素方差分析比較組間差異,P0.05為差異有統(tǒng)計(jì)學(xué)意義。 結(jié)果:1、;撬嶂委熃M癇性發(fā)作持續(xù)時(shí)間及發(fā)作分級較空白組及治療對照組降低。2、;撬嶂委熃M苔蘚纖維出芽情況較空白組及治療對照組評分低。 結(jié)論:;撬崮軠p低癲癇發(fā)作的持續(xù)時(shí)間及發(fā)作分級,并對致癇鼠海馬苔蘚纖維出芽有干預(yù)作用。

The epileptiform rats were randomly divided into three groups: blank group, taurine treatment group and treatment control group with 10 rats in each group. The taurine treatment group received intraperitoneal injection of 2g/kg taurine every day for 6 days. To observe the behavioral changes of epileptic seizures in taurine treatment group at different time points and to compare the duration and grade of epileptic seizures. Behavioral observation referred to the Racine seizure grading standard and made a behavioral observation table to observe the behavior of epileptic seizures in each group of rats after modeling.
