人源α-synuclein跨細(xì)胞轉(zhuǎn)運(yùn)特性及對神經(jīng)細(xì)胞突起的影響

【摘要】:目的確定人源α-synuclein是否能夠跨細(xì)胞轉(zhuǎn)運(yùn),過表達(dá)人源α-synuclein對神經(jīng)細(xì)胞突起有何影響。方法建立穩(wěn)定表達(dá)人源α-synuclein的N2a細(xì)胞株,通過接觸和非接觸兩種方法觀察人源α-synuclein的轉(zhuǎn)運(yùn)現(xiàn)象,通過測量神經(jīng)突起長度,評價人源α-synuclein過表達(dá)對神經(jīng)細(xì)胞突起生長的影響。結(jié)果接觸和非接觸兩種方法均能夠觀察到人源α-synuclein跨細(xì)胞轉(zhuǎn)運(yùn),但總體轉(zhuǎn)運(yùn)率低。過表達(dá)人源α-synuclein導(dǎo)致神經(jīng)細(xì)胞突起生長不良。結(jié)論人源α-synuclein跨細(xì)胞轉(zhuǎn)運(yùn)現(xiàn)象存在,轉(zhuǎn)運(yùn)可能受到多因素影響,效率較低。過表達(dá)人源α-synuclein蛋白對神經(jīng)突起有損傷作用。
[Abstract]:Aim to determine whether human 偽 -synuclein can be translocated across cells and how over-expression of human 偽 -synuclein affects neuronal processes. Methods N2a cell lines expressing human 偽 -synuclein stably were established. The transport of human 偽 -synuclein was observed by contact and non-contact methods. The effect of human 偽 -synuclein over-expression on neurite growth was evaluated by measuring the length of neurite. Results both contact and non-contact methods could observe the transcellular transport of human 偽 -synuclein, but the overall transport rate was low. Overexpression of human 偽-synuclein leads to poor neurite growth. Conclusion the transcellular transport of human 偽 -synuclein may be affected by many factors and its efficiency is low. Overexpression of human 偽-synuclein protein can damage the neurite.
