低頻重復(fù)經(jīng)顱磁刺激對卒中后抑郁大鼠行為學(xué)及海馬c-fos、c-jun蛋白表達(dá)的影響

【摘要】:目的:探討重復(fù)經(jīng)顱磁刺激(rTMS)對卒中后抑郁(PSD)模型大鼠行為學(xué)及海馬c-fos、c-jun蛋白表達(dá)的影響。方法:篩選40只健康雄性SD大鼠按隨機(jī)數(shù)字表法分為對照組、假手術(shù)組、模型組和治療組(n=10)。對照組不給予任何干預(yù)處理;模型組和治療組采用大腦中動脈栓塞法(MCAO)和慢性不可預(yù)見溫和應(yīng)激法(CUMS)建立PSD模型;假手術(shù)組和治療組給予rTMS治療;通過蔗糖水消耗實(shí)驗(yàn)、曠野實(shí)驗(yàn)和水迷宮實(shí)驗(yàn)對各組大鼠行為學(xué)進(jìn)行評估;應(yīng)用免疫組化實(shí)驗(yàn)檢測各組大鼠海馬c-fos、c-jun的陽性細(xì)胞灰度值。結(jié)果:(1)蔗糖水消耗實(shí)驗(yàn):與模型組比較,治療組蔗糖水消耗量明顯增多(P0.05);(2)曠野實(shí)驗(yàn):治療組大鼠水平及垂直得分較模型組有顯著提高(P0.05);(3)水迷宮實(shí)驗(yàn):治療組大鼠較模型組學(xué)習(xí)和記憶能力都有所改善(P0.05);(4)免疫組化實(shí)驗(yàn):治療組大鼠c-fos陽性細(xì)胞灰度值高于模型組(P0.05),各組間c-jun陽性細(xì)胞灰度值差異不顯著(P0.05)。結(jié)論:rTMS能夠有效改善PSD模型大鼠的抑郁樣行為,其機(jī)制可能與海馬c-fos、c-jun蛋白的表達(dá)有關(guān)。
[Abstract]:Aim: to investigate the effects of repeated transcranial magnetic stimulation (rTMS) on the behavior and expression of c-fossil-c-jun protein in hippocampus of poststroke depression (PSD) rats. Methods: forty male Sprague-Dawley rats were randomly divided into control group, sham operation group, model group and treatment group (n = 10). The model group and the treatment group were treated with middle cerebral artery embolization (MCAO) and chronic unpredictable mild stress (CUMS), the sham-operated group and the treatment group were treated with rTMS, and the sucrose water consumption test was used. Field experiment and water maze test were used to evaluate the behavior of rats in each group, and the gray scale of c-fos-c-jun positive cells in hippocampus of each group was detected by immunohistochemistry. Results: (1) Sucrose water consumption experiment: compared with the model group, The consumption of sucrose water in the treatment group was significantly increased (P0.05); (2). The scores of horizontal and vertical in the treatment group were significantly higher than those in the model group (P0.05); (3). The learning and memory abilities of the treatment group were improved compared with the model group (P0.05); (4). Immunohistochemical experiment: the gray value of c-fos positive cells in the treatment group was higher than that in the model group (P0.05), but there was no significant difference in the gray value of c-jun positive cells among the groups (P0.05). ConclusionTwo rTMS can effectively improve the depressive behavior of PSD model rats, and its mechanism may be related to the expression of c-fos-c-jun protein in hippocampus.
