全反式維甲酸對大鼠腦缺血再灌注損傷后MMP-9表達(dá)的影響

【摘要】:目的通過觀察全反式維甲酸(all trans retinoic acid,ATRA)對缺血再灌注大鼠基質(zhì)金屬蛋白酶-9(matrix metalloproteinase9, MMP-9)表達(dá)的影響,探討ATRA的神經(jīng)保護(hù)作用及可能機(jī)制。 方法健康雄性成年SD大鼠180只,隨機(jī)分為3組,每組60只,即假手術(shù)組(Sham組),缺血再灌注手術(shù)組(OP組)和ATRA干預(yù)組(ATRA組)。Sham組只行頸部手術(shù),不行血管栓塞,ATRA組和OP組按ZeaLonga方法行大腦中動脈栓塞。ATRA組術(shù)后每日予以30mg/kgATRA油溶液灌胃,Sham組和OP組每日食用油灌胃。每組分別于造模后1天,3天和7天,行TTC染色、EB染色、免疫組化和Western Blot檢測MMP-9。 結(jié)果 1.神經(jīng)功能評分 Sham組大鼠無運(yùn)動功能缺損,Bederson評分均為0分。與OP組比較,ATRA3天組大鼠Bederson評分有顯著改善(P0.05),1天組和7天組改善不顯著(P0.05)。 2.TTC染色 Sham組大鼠TTC染色未發(fā)現(xiàn)梗死區(qū)域。OP組1天時(shí)腦梗死最明顯,3天、7天時(shí)梗死體積逐漸減;和OP組比較,ATRA組各時(shí)間點(diǎn)梗死體積均減小,其中1天、7天組有統(tǒng)計(jì)學(xué)意義(P0.05)。 3.EB含量 Sham組各時(shí)間點(diǎn)腦組織EB含量均較低。OP組EB含量再灌注后1天達(dá)高峰,后逐漸下降,7天尚未降至Sham組水平。與Sham組相比,OP組大鼠1天、3天、7天各時(shí)間點(diǎn)EB含量均顯著升高(P0.05)。與OP組相比,ATRA組各時(shí)間點(diǎn)EB含量均減少,其中1天組和7天組具有統(tǒng)計(jì)學(xué)意義(P0.05)。 4.免疫組化 Sham組大鼠腦組織內(nèi)基本無MMP-9表達(dá)。OP組和ATRA組大鼠MMP-9主要在梗死區(qū)及梗死區(qū)附近的膠質(zhì)細(xì)胞的胞漿中表達(dá)。與Sham組相比,OP組MMP-9的表達(dá)明顯增多,其中3天,7天組具有統(tǒng)計(jì)學(xué)意義(P0.05)。與OP組比較,ATRA組各時(shí)間點(diǎn)MMP-9的表達(dá)均減少,其中3天和7天組有統(tǒng)計(jì)學(xué)意義(P0.05)。 5.Western Blot 與Sham組比較,OP組1天、3天、7天各時(shí)間點(diǎn)MMP-9的表達(dá)均顯著增加(P0.05)。與OP組比較,ATRA組1天、3天和7天組MMP-9表達(dá)均顯著降低(P0.05)。 結(jié)論 ATRA減輕MCAO大鼠血腦屏障的損傷,可能機(jī)制為通過下調(diào)MMP-9的表達(dá)。
[Abstract]:Objective To investigate the effects of all trans retinoic acid ( ATRA ) on the expression of matrix metalloproteinase - 9 ( MMP - 9 ) in ischemia - reperfusion rats .
Methods 180 healthy male adult SD rats were randomly divided into three groups : sham operation group ( Sham group ) , ischemia / reperfusion group ( OP group ) and ATRA group ( ATRA group ) . The Sham group only underwent cervical operation , no vascular embolism , ATRA group and OP group were injected into the middle cerebral artery embolization according to ZeaLonga method . All groups were treated with 30 mg / kg ATRA oil solution for 1 day , 3 days and 7 days , TTC staining , EB staining , immunohistochemistry and Western Blot were used to detect MMP - 9 .
Results
1 . Nerve function score
Compared with OP group , the score of Bederson ' s score in ATRA3 - day group was significantly improved ( P0.05 ) , and the improvement of group 1 - day group and 7 - day group was not significant ( P0.05 ) .
2 . TTC Staining
The infarct area was not found in the control group of the Sham group . The cerebral infarction was the most obvious in OP group at 1 day , and the infarct volume was gradually decreased in 3 days and 7 days .
Compared with OP group , the infarct volume decreased at all time points of ATRA group , among which 1 day and 7 day group had statistical significance ( P0.05 ) .
3.EB Content
Compared with Sham group , the EB content of all time points in OP group increased significantly ( P0.05 ) . Compared with the Sham group , the EB content in all time points in OP group decreased significantly ( P0.05 ) .
4 . Immunohistochemical technique
Compared with Sham group , the expression of MMP - 9 in OP group was significantly higher than that in Sham group ( P0.05 ) .
5.Western Blot
Compared with Sham group , the expression of MMP - 9 was significantly increased in OP group 1 day , 3 day and 7 days ( P0.05 ) . Compared with OP group , the expression of MMP - 9 in group 1 , 3 and 7 days in ATRA group was significantly decreased ( P0.05 ) .
Conclusion
ATRA reduces the damage of blood - brain barrier in the rats with middle cerebral artery occlusion , which may be mediated by downregulating the expression of MMP - 9 .
