膠質(zhì)瘤中SPINT2基因的表達及其甲基化狀態(tài)研究

【摘要】:目的:檢測正常腦組織標本及不同WHO分級的人腦膠質(zhì)瘤標本中SPINT2基因的表達及其甲基化狀態(tài),探討SPINT2基因甲基化對膠質(zhì)瘤發(fā)生、發(fā)展過程的影響。 方法:選取8例正常腦組織及24例不同病理等級的膠質(zhì)瘤組織,用western blot方法檢測各標本中SPINT2基因表達情況;同時使用甲基化特異性PCR(MSP)檢測SPINT2基因在正常腦組織和膠質(zhì)瘤中啟動子甲基化情況。 結(jié)果:1.Western-blot檢測正常腦組織、膠質(zhì)瘤組織中SPINT2蛋白表達實驗結(jié)果:正常腦組織中SPINT2蛋白與低級別膠質(zhì)瘤差異具有統(tǒng)計學(xué)意義(P=0.008);正常腦組織中SPINT2蛋白與高級別膠質(zhì)瘤差異具有統(tǒng)計學(xué)意義(P=0.000);低級別膠質(zhì)瘤中SPINT2蛋白與高級別膠質(zhì)瘤差異無統(tǒng)計學(xué)意義(P=0.061)。2. MSP檢測正常腦組織、膠質(zhì)瘤組織中SPINT2基因甲基化狀態(tài)實驗結(jié)果:正常腦組織中SPINT2基因啟動子甲基化率與低級別膠質(zhì)瘤差異無統(tǒng)計學(xué)意義(P=0.347);正常腦組織中SPINT2基因啟動子甲基化率與高級別膠質(zhì)瘤差異具有統(tǒng)計學(xué)意義(P=0.024);低級別膠質(zhì)瘤中SPINT2基因啟動子甲基化率與高級別膠質(zhì)瘤差異無統(tǒng)計學(xué)意義(P=0.097)。 結(jié)論:1.SPINT2基因在所有正常腦組織及大部分膠質(zhì)瘤組織中均可表達;正常腦組織中SPINT2蛋白含量明顯高于膠質(zhì)瘤組織;低級別膠質(zhì)瘤組織中SPINT2蛋白含量與高級別膠質(zhì)瘤組織相比無明顯差異。2.正常腦組織中未發(fā)現(xiàn)SPINT2基因啟動子出現(xiàn)甲基化現(xiàn)象;正常腦組織中SPINT2基因啟動子甲基化率明顯低于高級別膠質(zhì)瘤組織;而正常腦組織中SPINT2基因啟動子甲基化率與低級別膠質(zhì)瘤組織、低級別膠質(zhì)瘤組織中SPINT2基因啟動子甲基化率與高級別膠質(zhì)瘤組織相比無明顯差異。圖7幅,表6個,參考文獻32篇。
[Abstract]:Aim: to detect the expression and methylation status of SPINT2 gene in normal brain tissues and human gliomas with different WHO grades, and to investigate the effect of methylation of SPINT2 gene on the occurrence and development of gliomas.Methods: the expression of SPINT2 gene in 8 normal brain tissues and 24 gliomas with different pathological grades was detected by western blot method.The promoter methylation of SPINT2 gene in normal brain tissue and glioma was detected by methylation specific PCR MSPs.Results 1. Western-blot was used to detect normal brain tissue.The expression of SPINT2 protein in normal brain tissue was significantly different from that in low-grade glioma (P < 0.008), SPINT2 protein in normal brain tissue was significantly higher than that in high-grade glioma (P = 0.000), and low grade glioma was lower grade (P < 0.05).There was no significant difference between SPINT2 protein and high grade glioma.MSP was used to detect normal brain tissue.Results of methylation status test of SPINT2 gene in glioma tissues: the methylation rate of SPINT2 gene promoter in normal brain tissue was not significantly different from that in low-grade glioma tissue, and the methylation rate of SPINT2 gene promoter in normal brain tissue was higher than that in low-grade glioma tissue.The methylation rate of SPINT2 gene promoter in low grade gliomas was not significantly different from that in high grade gliomas.Conclusion: 1. SPINT2 gene can be expressed in all normal brain tissues and most glioma tissues, and the content of SPINT2 protein in normal brain tissue is significantly higher than that in glioma tissue.There was no significant difference in SPINT2 protein content between low grade gliomas and high grade gliomas.No methylation of SPINT2 gene promoter was found in normal brain tissue, and the methylation rate of SPINT2 gene promoter in normal brain tissue was significantly lower than that in high grade glioma tissue.However, the methylation rate of SPINT2 gene promoter in normal brain tissue was not significantly different from that in low-grade glioma tissue and low-grade glioma tissue, and the methylation rate of SPINT2 gene promoter in low-grade glioma tissue was not significantly different from that in high-grade glioma tissue.There are 7 figures, 6 tables and 32 references.
