BAFF和BAFFR在難治性癲癇患者及大鼠模型中的表達(dá)變化

【摘要】:目的:BAFF主要通過調(diào)控BAFFR受體參與中樞神經(jīng)系統(tǒng)免疫炎癥反應(yīng)的病理生理過程。目前尚不清楚BAFF和BAFFR是否也參與了癲癇的病理過程中。本研究主要通過對難治性癲癇患者以及癲癇大鼠模型腦組織中BAFF和BAFFR的表達(dá)變化,探討B(tài)AFF和BAFFR在癲癇發(fā)病機(jī)制的作用。方法:1.本實驗以24例難治性癲癇患者手術(shù)切除的顳葉病灶標(biāo)本為實驗組,以8例腦外傷患者手術(shù)切除的顳葉皮質(zhì)作為對照組。2.將30只SD雄性大鼠隨機(jī)均分為實驗組和對照組,實驗組SD大鼠給予匹魯卡品腹腔注射,對照組給予生理鹽水腹腔注射,觀察癲癇動物模型的急性發(fā)作情況及2個月后的慢性發(fā)作情況。3.采用免疫熒光和免疫組化對BAFF和BAFFR進(jìn)行蛋白表達(dá)定位,Western-blot和RT-qPCR技術(shù)對BAFF和BAFFR進(jìn)行到蛋白表達(dá)定量。結(jié)果:免疫熒光結(jié)果顯示BAFF和BAFFR在癲癇患者和大鼠模型神經(jīng)元的細(xì)胞質(zhì)和細(xì)胞膜中表達(dá),在神經(jīng)膠質(zhì)細(xì)胞中不表達(dá);免疫組化結(jié)果顯示BAFF和BAFFR蛋白在癲癇患者及動物模型神經(jīng)元的細(xì)胞質(zhì)和細(xì)胞膜中都有表達(dá),并且表達(dá)強(qiáng)度明顯高于對照組(P0.05);Western-blot和RT-PCR結(jié)果顯示BAFF和BAFFR在癲癇患者及動物模型中表達(dá)量皆較對照組明顯升高(P0.05)。結(jié)論:BAFF和BAFFR在癲癇患者及癲癇大鼠模型中表達(dá)量明顯升高。目前研究表明BAFF和BAFFR參與中樞神經(jīng)系統(tǒng)的免疫及炎癥反應(yīng),故我們推測升高的BAFF和BAFFR是通過免疫炎癥反應(yīng)來調(diào)節(jié)癲癇的發(fā)病過程并具有一定的作用。
[Abstract]:Objective: BAFF mainly through the pathophysiological process of regulation of BAFFR receptor in the central nervous system immune inflammation. It is unclear whether BAFF and BAFFR are involved in the pathophysiology of epilepsy. Through the research on the expression of patients with intractable epilepsy and epilepsy rat model of brain tissue of BAFF and BAFFR, to explore the the role of BAFF and BAFFR in the pathogenesis of epilepsy. Methods: 1. in this experiment, 24 cases of refractory epilepsy surgery in patients with temporal lobe lesions were chosen as experimental group, 8 patients with temporal lobe resection in patients with traumatic brain injury as the control group.2. 30 SD male rats were randomly divided into experimental group and control group, the experimental group SD rats were given intraperitoneal injection of pilocarpine, the control group was given intraperitoneal injection of normal saline, acute attack were observed in animal model of epilepsy and 2 months after the onset of chronic.3. by immunofluorescence and free Immunohistochemistry of BAFF and BAFFR protein expression, Western-blot and RT-qPCR technology of BAFF and BAFFR protein expression. Results: quantitative immunofluorescence showed that the expression of BAFF and BAFFR in neurons of rat model of epilepsy and the cytoplasm and cell membrane, no expression in glial cells; immunohistochemistry BAFF and BAFFR protein were expressed in neurons of epileptic patients and animal models of the cytoplasm and cell membrane, and the expression intensity was significantly higher than the control group (P0.05); Western-blot and RT-PCR showed that BAFF and BAFFR were significantly higher than the control group expression in epileptic patients and animal models (P0.05). Conclusion: the expression of BAFF and BAFFR in patients with epilepsy and epilepsy model in rats increased significantly. The present study shows that immune and inflammatory reaction of BAFF and BAFFR in the central nervous system, so we speculate that the rise of BAFF And BAFFR can regulate the process of epilepsy by immuno inflammatory response and play a role in the pathogenesis of epilepsy.
