不同程度彌漫性軸索損傷動物模型建立研究

【摘要】:目的自制一種彌漫性軸索損傷(DAI)裝置,建立不同程度的DAI動物模型。方法選取成年雄性清潔級SD大鼠70只,分為正常對照組(10只)與致傷組(60只),進(jìn)一步將致傷組分為A、B、C 3組,各20只。運用自制的實驗裝置致傷,使大鼠的頭顱同時產(chǎn)生瞬間超大角加速度和線加速度,在恒定氣壓下通過不同的旋轉(zhuǎn)往返加速運動次數(shù),造成輕型(A組)、中型(B組)、重型(C組)3種不同程度的大鼠DAI損傷模型,進(jìn)行行為及病理效應(yīng)評價。結(jié)果隨著損傷程度的加重,損傷急性期致傷組大鼠的神經(jīng)生理反射恢復(fù)的時間間隔與蘇醒時間均增加(P0.05),致傷7d后致傷組大鼠的神經(jīng)功能評分降低(P0.05);致傷后14d內(nèi)A、B、C 3組大鼠死亡率分別為5.0%、25.0%、50.0%。隨著損傷程度的加重,DAI病理特征改變更為明顯。結(jié)論該裝置能有效建立不同損傷程度的DAI動物模型。
[Abstract]:Objective to establish the animal model of diffuse axonal injury (DAI) by self-made (DAI) device. Methods 70 adult male SD rats were divided into normal control group (n = 10) and injury group (n = 60). The injured rats were further divided into A, B and C groups with 20 rats in each group. The injury was caused by a self-made experimental device, which made the rat's head produce instantaneous super-angular acceleration and linear acceleration at the same time, and accelerated the movement through different rotations under constant air pressure, resulting in light weight (group A) and medium-sized (group B). Severe (group C) three different degrees of DAI damage models were evaluated for their behavior and pathological effects. Results with the severity of injury, the interval of recovery of neurophysiological reflex and recovery time of rats in acute stage of injury increased (P0.05), and the neurological function score of injured group decreased 7 days after injury (P0.05). Within 14 days after injury, the mortality rates of A, B and C groups were 5.0%, 25.0% and 50.0%, respectively. With the severity of injury, the pathological changes of DAI were more obvious. Conclusion this device can effectively establish the animal model of DAI with different degree of injury.
