殼聚糖蜂蠟膏對大鼠創(chuàng)面中CD34及羥脯氨酸的影響

【摘要】:目的觀察殼聚糖蜂蠟膏對大鼠創(chuàng)面中CD34、羥脯氨酸的影響及創(chuàng)面愈合效果并探討其可能機制。 方法取20只雄性SD大鼠,在其背部兩側制造全層皮膚缺損創(chuàng)面,制成創(chuàng)面模型,每只大鼠背部三個創(chuàng)面隨機分為實驗組、對照組和空白對照組。造模后每隔2d換藥1次,空白對照組不做處理,實驗組創(chuàng)面使用5g殼聚糖蜂蠟膏,對照組使用5ml生理鹽水。免疫組化法評價創(chuàng)面組織中CD34表達情況,計數創(chuàng)面中新生毛細血管數量;酸水解法檢測羥脯氨酸在創(chuàng)面組織中的表達;觀察創(chuàng)面的未愈合面積、創(chuàng)面愈合率;計量資料用均數±標準差(X±s)表示,采用SPSS13.0統(tǒng)計學軟件。兩樣本均數比較應用配對t檢驗,率的比較應用卡方檢驗。P0.05為差異具有統(tǒng)計學意義。 結果實驗組第7、14、21天時創(chuàng)面組織中CD34表達量高于同期對照組;第7天時實驗組毛細血管計數高于對照組,但無統(tǒng)計學差異(P0.05),第14、21天時實驗組毛細血管數均高于同期對照組(表1),有統(tǒng)計學差異(P0.05)。實驗組第7、14、21天時創(chuàng)面組織中羥脯氨酸含量均高于同期對照組(表2),差異有統(tǒng)計學意義(P0.05)。第7天實驗組創(chuàng)面愈合率大于對照組,但無統(tǒng)計學差異(P0.05);第14d、21d實驗組創(chuàng)面愈合率高于同期對照組(表3),具有統(tǒng)計學意義(P0.05)。 結論殼聚糖蜂蠟膏能促進創(chuàng)面組織中CD34、羥脯氨酸表達及毛細血管再生,可能是促進創(chuàng)面愈合的機制之一。
[Abstract]:Objective to observe the effect of chitosan beeswax ointment on CD34, hydroxyproline in rat wound and to explore its possible mechanism. Methods 20 male SD rats were used to make full-thickness skin defect wounds on both sides of their backs. Each rat was randomly divided into experimental group, control group and blank control group. Every 2 days, the blank control group was not treated, the experimental group was treated with 5 g chitosan beeswax ointment, and the control group was treated with 5ml saline. Immunohistochemical method was used to evaluate the expression of CD34 in wound tissue and count the number of neocapillaries in the wound, acid hydrolysis method was used to detect the expression of hydroxyproline in the wound tissue, and the unhealed area and wound healing rate of the wound were observed. The measurement data were expressed by mean 鹵standard deviation (X 鹵s), and SPSS13.0 software was used. The average of two samples was compared by paired t test, and the rate was compared by chi-square test. P0.05 was statistically significant. Results the expression of CD34 in the wound tissue of the experimental group was higher than that of the control group at day 7 and day 14. On the 7th day, the capillary count in the experimental group was higher than that in the control group, but there was no statistical difference (P0.05). On the 14th day, the capillary count in the experimental group was higher than that in the control group (table 1), and there was statistical difference (P0.05). The content of hydroxyproline in the wound tissue of the experimental group was significantly higher than that of the control group (P 0.05). On the 7th day, the wound healing rate in the experimental group was higher than that in the control group, but there was no statistical difference (P0.05); on the 14th day, the wound healing rate in the experimental group was higher than that in the control group (table 3), which had statistical significance (P0.05). Conclusion chitosan beeswax ointment can promote the expression of CD34, hydroxyproline and capillary regeneration in wound tissue, which may be one of the mechanisms of promoting wound healing.
