HBV相關(guān)ACLF患者PBMCs中ROG、GATA3和T-bet mRNA水平檢測

【摘要】:目的檢測HBV相關(guān)慢加急性肝衰竭(ACLF)患者外周血單個核細(xì)胞(PBMC)中GATA3抑制因子(ROG)、GATA3和T-bet mRNA水平變化及意義。方法收集45例HBV相關(guān)ACLF(HBV-ACLF)患者和15例健康志愿者的外周血,分離單個核細(xì)胞,提取總RNA,用實時定量PCR的方法檢測ROG、GATA3和T-bet mRNA水平。結(jié)果 HBV-ACLF患者和健康對照ROG水平分別為0.98±0.27和0.64±0.19,差異有統(tǒng)計學(xué)意義(P0.05);HBV-ACLF患者和健康對照GATA3水平分別為0.73±0.15和0.58±0.22,差異有統(tǒng)計學(xué)意義(P0.05);HBV-ACLF患者和健康對照T-bet水平分別為0.78±0.29和0.46±0.17,差異有統(tǒng)計學(xué)意義(P0.05);HBV-ACLF患者和健康對照ROG/GATA3和T-bet/GATA3比較,差異均有統(tǒng)計學(xué)意義(P0.05)。HBVACLF患者ROG表達(dá)水平與GATA3 mRNA呈負(fù)相關(guān)(r=-0.724,P0.05)。而ROG mRNA表達(dá)水平與T-bet/GATA3比值無相關(guān)性(r=-0.064,P0.05)。結(jié)論 ROG、GATA3和T-bet在HBV-ACLF患者PBMC中表達(dá)水平上調(diào)明顯,參與肝功能衰竭的發(fā)病發(fā)生和發(fā)展。
[Abstract]:Objective to investigate the changes and significance of GATA3 inhibitory factor (GATA3) in peripheral blood mononuclear cells (PBMCs) of patients with HBV associated with chronic and acute hepatic failure (ACLF).Methods Peripheral blood samples were collected from 45 patients with HBV associated with HBV-ACLF and 15 healthy volunteers. Mononuclear cells were isolated, total RNAs were extracted, and the levels of ROGN GATA3 and T-bet mRNA were detected by real-time quantitative PCR.There were significant differences in ROG/GATA3 and T-bet/GATA3 between patients with HBV-ACLF and healthy controls.There was a negative correlation between the expression of ROG and GATA3 mRNA in patients with P0.05. HBV-ACLF, and there was a negative correlation between the expression of ROG and GATA3 mRNA (P 0.05).However, there was no correlation between the expression of ROG mRNA and the ratio of T-bet/GATA3.Conclusion the expression of Gata _ 3 and T-bet in PBMC of HBV-ACLF patients is up-regulated, which is involved in the pathogenesis and development of liver failure.
