伊伐布雷定對急性心肌梗死兔血清中低氧誘導因子-1α和VEGF表達的影響

【摘要】:目的:探討伊伐布雷定對急性心肌梗死模型兔血清中低氧誘導因子-1α(HIF-1a)和血管內皮生長因子(VEGF)表達的影響。 方法:2]只新西蘭大白兔隨機分為假手術組(S)、心肌梗死組(M)、富馬酸比索洛爾治療組(M+B)、伊伐布雷定治療組(M+I),其中M+B組自心肌梗死模型當天起以富馬酸比索洛爾1.25mg每天1次,服藥1周后再以2.5mg每天1次繼續(xù)服藥2周,M+I組以伊伐布雷定17mg/(kg·d)服藥3周,對各組兔分別于術前24h、術后24h、術后3d、術后1周、術后2周、術后3周,6個時間點自耳緣靜脈抽取靜脈血,采用Elisa法檢測血清中HIF-1a和VEGF水平。采用心電圖機記錄并分析各組兔術前及術后3周的心率變化。 結果:3組心肌梗死兔基礎心率比較無統計學差異;術后3周與M組相比,M+B和M+I兩組心率明顯減慢(F=49.61,q=7.54,,P0.05),而M+B和M+I兩組相比心率減慢程度無明顯差異(P0.05); M、M+B、M+I組不同時間點血清中HIF-1a和VEGF水平與S組相比均明顯升高,差異有顯著性(F=8.22-11.88, q=4.32-6.73, P0.05), M+B、M+I兩組較M組明顯降低(F=5.23-12.11, q=3.97-8.32, P0.05), M+B、M+I兩組比較差異無顯著性(P0.05);各組兔血清中HIF-1a與VEGF水平呈正相關性(r=0.732,P0.01)。 結論:伊伐布雷定在減慢心率的同時,可降低心肌梗死后血清中HIF-1a和VEGF水平,其療效與比索洛爾無明顯差異。
[Abstract]:Aim: to investigate the effects of ivalburetine on the expression of hypoxia inducible factor 1 偽 (HIF-1a) and vascular endothelial growth factor (VEGF) in serum of rabbits with acute myocardial infarction (AMI).Methods Twenty two New Zealand white rabbits were randomly divided into three groups: sham-operated group, myocardial infarction group, bisoprolol fumarate group, and ivalburetine treatment group. The M B group was treated with 1.25mg once a day since the myocardial infarction model.After 1 week of administration, 2.5mg was given once a day for 2 weeks. The rabbits in the MI group were treated with 17mg/(kg d for 3 weeks. The rabbits in each group were treated at 24 hours before operation, 24 hours after operation, 3 days after operation, 1 week after operation and 2 weeks after operation, respectively.At 3 weeks after operation, venous blood was extracted from the auricular vein at 6 time points and the levels of HIF-1a and VEGF in serum were detected by Elisa method.The changes of heart rate before and 3 weeks after operation were recorded and analyzed by electrocardiograph.Results there was no significant difference in basal heart rate among the three groups.鏈悗3鍛ㄤ笌M緇勭浉姣
