【摘要】：研究目的 1、觀察載脂蛋白E(ApoE)模擬肽ApoE23對膿毒血癥小鼠血漿LPS濃度變化的影響。 2、研究ApoE23對膿毒血癥小鼠肝臟LPS代謝相關受體表達的調(diào)節(jié)作用。 研究方法 1、ApoE23合成采用固相合成法,高效液相色譜(HPLC)技術進行純化；采用B組鼠傷寒沙門菌誘導膿毒血癥C57BL小鼠模型；實驗小鼠分三組：(1)感染對照組；(2)膿毒血癥組；(3) ApoE23治療組。 2、不同組小鼠血漿LPS(磷酸脂多糖)濃度測定采用連續(xù)透射免疫比濁法。 3、采用定量RT-PCR技術及Western-Blot技術檢測不同組別小鼠肝臟中低密度脂蛋白受體(LDLR)、低密度脂蛋白受體相關蛋白(LRP)、多配體蛋白聚糖-1(SDC1)、清道夫受體B1(SRB1)及ApoEmRNA及蛋白質表達水平的變化。 研究結果 1、膿毒血癥組小鼠腹腔注射B組鼠傷寒沙門菌后1h血液細菌培養(yǎng)即分離到B組鼠傷寒沙門氏菌,小鼠肝臟病理切片表現(xiàn)為不同程度的炎性細胞浸潤。血漿LPS濃度測定發(fā)現(xiàn),細菌腹腔注射后1h,小鼠血漿中LPS濃度已開始升高,3h、24h維持在較高水平。ApoE23治療后小鼠肝臟炎癥病變明顯減輕,血漿LPS濃度也明顯低于膿毒血癥組。 2、與感染對照組相比,膿毒血癥組小鼠肝臟LDLR、LRP、SDC1在mRNA和蛋白質表達水平均有不同程度的降低,而SRB1變現(xiàn)為不同程度的升高；與膿毒血癥組相比,ApoE23治療后,肝臟LDLR在1、3、24h的mRNA和蛋白質表達水平均升高；LRP在mRNA表達水平?jīng)]有改變,1h和3h蛋白質表達水平升高；SDC1與SRB1在1h的mRNA表達水平表現(xiàn)為一過性變化。 3、與感染對照組相比,膿毒血癥組小鼠肝臟ApoE在1h、3h的mRNA和蛋白質水平表達下降；ApoE23治療后,與膿毒血癥組相比,ApoE在mRNA和蛋白質表達水平均沒有明顯改變。 結論 1、ApoE23能夠明顯改善膿毒血癥小鼠肝臟組織炎癥侵潤,并明顯降低膿毒血癥小鼠血漿中LPS濃度,發(fā)揮抗膿毒血癥效應。 2、膿毒血癥時肝臟LPS代謝相關受體表達下降,導致LPS在肝臟中代謝障礙,上述病理生理變化加重血漿LPS濃度的升高。ApoE23治療促進肝臟LDLR, LRP表達水平回升,加速LPS在肝臟中的代謝,發(fā)揮抗膿毒血癥效應。 3、ApoE對膿毒血癥的治療作用目前仍存在爭議,ApoE缺陷或是濃度過高均有可能誘發(fā)膿毒血癥,所以膿毒血癥時保持穩(wěn)定的血漿ApoE水平將有利于膿毒血癥的轉歸。由于ApoE主要在肝臟合成及代謝,藥物ApoE23治療之后我們發(fā)現(xiàn)肝臟ApoE的表達沒有發(fā)生改變,外源性ApoE23不會對肝臟本身ApoE的合成代謝產(chǎn)生影響。
[Abstract]:Research purpose. 1. The effect of apolipoprotein (ApoE) mimic peptide ApoE23 on plasma LPS concentration in sepsis mice was observed. 2. To study the regulatory effect of ApoE23 on the expression of LPS metabolism-related receptors in the liver of sepsis mice. Research method. (1) the synthesis of ApoE23 was purified by high performance liquid chromatography (HPLC) and purified by solid phase synthesis, the sepsis C57BL mice model was induced by Salmonella typhimurium in group B, and the experimental mice were divided into three groups: control group (control group), control group (n = 2), sepsis group (n = 3) ApoE23 treatment group. 2. Plasma LPS (lipopolysaccharide) concentration in different groups was determined by continuous transmission immunoturbidimetry. 3. Quantitative RT-PCR and Western-Blot techniques were used to detect the expression levels of LDLRN, LRPX, SDC1, B1 SDC1, ApoEmRNA and protein in the liver of different groups of mice. Research results. 1. Salmonella typhimurium was isolated from the blood of mice in group B after intraperitoneal injection of Salmonella typhimurium in group B. the pathological sections of the liver of the mice showed inflammatory cell infiltration in varying degrees. The plasma LPS concentration was measured. 1 h after intraperitoneal injection of bacteria, the plasma LPS concentration in mice began to increase and maintained at a higher level for 24 h. After treatment with ApoE23, the inflammatory lesion of liver was significantly alleviated, and the plasma LPS concentration was significantly lower than that in sepsis group. 2. Compared with the infected control group, the expression of LDLRP-SDC1 in liver of sepsis group was decreased in different degree, but SRB1 was increased in different degree, and compared with that in sepsis group, the expression level of LDLRP-SDC1 in liver of sepsis group was higher than that of sepsis group, and the expression of LRP-SDC1 in liver of sepsis group was higher than that of sepsis group. The expression level of mRNA and protein in liver LDLR increased at 1h and 3h. The expression level of mRNA and protein in mRNA did not change at 1h and 3h. The mRNA expression of SDC1 and SRB1 at 1h showed a transient change. 3Compared with the infected control group, the expression of mRNA and protein in the liver of sepsis group decreased at 1h and 3h after treatment with ApoE23, but the expression level of mRNA and protein in the sepsis group was not significantly changed compared with that in the sepsis group. Conclusion. 1 ApoE23 could obviously improve the inflammation invasion of liver tissue in sepsis mice, decrease the concentration of LPS in plasma of sepsis mice, and play the role of anti-sepsis effect. 2. The expression of LPS metabolism-related receptors in the liver decreased during sepsis, which resulted in the dysfunction of LPS metabolism in the liver. The above pathophysiological changes aggravated the increase of plasma LPS concentration. ApoE23 treatment promoted the increase of liver LDLR, LRP expression, and accelerated the metabolism of LPS in the liver. Give play to the effect of antisepsis. (3) the therapeutic effect of ApoE on sepsis is still controversial. The deficiency of ApoE or the excessive concentration of ApoE may induce sepsis. Therefore, maintaining stable plasma ApoE levels during sepsis is conducive to the outcome of sepsis. Since ApoE is mainly synthesized and metabolized in the liver, we found that the expression of ApoE in the liver did not change after the treatment of the drug ApoE23. Exogenous ApoE23 has no effect on the synthesis and metabolism of ApoE in liver itself.
