腦源性神經(jīng)營(yíng)養(yǎng)因子預(yù)處理神經(jīng)干細(xì)胞移植對(duì)急性缺血性腦卒中小鼠的效果

【摘要】:目的比較腦源性神經(jīng)營(yíng)養(yǎng)因子(BDNF)預(yù)處理對(duì)神經(jīng)干細(xì)胞(NSCs)移植修復(fù)急性缺血性腦卒中的作用。方法出生1 d C57BL/6小鼠,分離NSCs體外培養(yǎng)及鑒定。10周齡健康C57BL/6小鼠150只隨機(jī)分為5組,A組(n=20)為假手術(shù)組,B(n=20)、C(n=20)、D(n=45)、E(n=45)組采用光化學(xué)誘導(dǎo)法建立急性腦缺血模型。造模24 h后,D組行單純NSCs移植,E組行BDNF預(yù)處理的NSCs移植,C組移植等量溶劑。移植前1 d,移植后3 d、7 d、14 d、21 d、28 d,行加速轉(zhuǎn)棒測(cè)試及前肢抓力測(cè)試,移植后28 d,D、E組取5只小鼠行微管相關(guān)蛋白2(MAP-2)、神經(jīng)膠質(zhì)纖維酸性蛋白(GFAP)免疫熒光染色。結(jié)果移植后3 d、7 d、14d、21 d、28 d,加速轉(zhuǎn)棒時(shí)間由長(zhǎng)到短依次為E組、D組、B組(P0.05);移植后14 d、21 d、28 d,前肢抓力由大到小依次為E組、D組、B組(P0.05)。移植后28 d,D組、E組均發(fā)現(xiàn)Edu/GFAP雙陽(yáng)性細(xì)胞及Edu/MAP-2雙陽(yáng)性細(xì)胞。結(jié)論 NSCs移植可促進(jìn)缺血性腦卒中后行為功能恢復(fù),經(jīng)BDNF預(yù)處理后有更好效果。
[Abstract]:Objective to compare the effects of brain-derived neurotrophic factor (BDNF) preconditioning on neural stem cell transplantation (NSCs) in the repair of acute ischemic stroke. Methods one day C57BL / 6 mice were born. A total of 150 healthy 10-week-old C57BL / 6 mice were isolated and identified in vitro. They were randomly divided into 5 groups (group A) as sham-operation group (group B) as sham-operation group (group B). Acute cerebral ischemia model was established in group D by photochemical induction. Group D was treated with simple NSCs transplantation after 24 hours of modeling. BDNF pretreated NSCs transplantation group C was treated with the same amount of solvent. 1 day before transplantation, 3 days after transplantation, 7 days after transplantation and 14 days after transplantation for 21 days and 28 days after transplantation, accelerated rotation test and forelimb grip test were performed. On the 28th day after transplantation, 5 mice in group E received immunofluorescence staining of microtubule-associated protein 2- MAP-2 and glial fibrillary acidic protein (GFAP). Results after 3 days of transplantation, 7 days after transplantation, 14 days after transplantation, 21 days after transplantation, 28 days after transplantation, the accelerated rotation time was in turn from group E to group D, group B, P0.05. After 14 days, 21 days and 28 days, the order of forelimb grasping force was E group D group and B group P 0.05. Both Edu/GFAP positive cells and Edu/MAP-2 double positive cells were found in E group 28 days after transplantation. Conclusion NSCs transplantation can promote the recovery of behavioral function after ischemic stroke. After pretreatment with BDNF, the effect is better.
