阿魏酸在慢性創(chuàng)面中的治療作用研究

【摘要】:目的:研究阿魏酸對(duì)糖尿病大鼠難愈性創(chuàng)面及兔耳缺血性難愈性創(chuàng)面是否有促愈合作用及可能機(jī)制。方法:20只雄性SD大鼠設(shè)為正常對(duì)照組,80只經(jīng)高糖高脂飼料喂養(yǎng)聯(lián)合45mg/kg鏈脲佐菌素腹腔注射誘導(dǎo)的糖尿病大鼠隨機(jī)平均分為4組:糖尿病對(duì)照組、凡士林涂抹組、5%阿魏酸涂抹組、20mg/kg阿魏酸灌胃組。大鼠鼠背制作2cm×2cm全層皮膚缺損創(chuàng)面,每日創(chuàng)面換藥并給藥干預(yù),正常對(duì)照組及糖尿病對(duì)照組僅換藥處理。40只雄性新西蘭大耳白兔,隨機(jī)平均分為4組:凡士林組、缺血對(duì)照組、5%阿魏酸組、貝復(fù)濟(jì)(rb-bFGF)組;兔耳經(jīng)缺血處理并驗(yàn)證缺血效果后制作兔耳腹側(cè)創(chuàng)面,每日創(chuàng)面換藥并給藥干預(yù),缺血對(duì)照組僅換藥處理。兩個(gè)動(dòng)物實(shí)驗(yàn)均于術(shù)后1、5、8、11、14、17天拍照觀察,記錄各組創(chuàng)面愈合情況并取材,計(jì)算創(chuàng)面各時(shí)間點(diǎn)愈合率,比較各組創(chuàng)面愈合時(shí)間。取材時(shí)將標(biāo)本由中線一分為二,一半固定后行蘇木精-伊紅染色觀察創(chuàng)面愈合情況及免疫組織化學(xué)染色顯示組織中血管內(nèi)皮生長(zhǎng)因子(Vascular endothelial growth factor.VEGF)、轉(zhuǎn)化生長(zhǎng)因子-β1(transforming growth factor-beta 1.TGFβ1)、血小板-內(nèi)皮細(xì)胞粘附分子(Platelet endothelial cell adhesion molecule-1,PECAM-1/CD31)的表達(dá)情況,一半用于提取RNA后PCR檢測(cè)VEGF、TGFβ1、缺氧誘導(dǎo)因子-1α(hypoxia inducible factor-1α.HIF-1α)的表達(dá)。結(jié)果:大鼠實(shí)驗(yàn)結(jié)果:大體觀察、各組大鼠各時(shí)間點(diǎn)創(chuàng)面愈合率、各組大鼠創(chuàng)面愈合時(shí)間及造創(chuàng)后8天各組創(chuàng)面組織HE染色均表明5%阿魏酸組及20mg/kg阿魏酸灌胃組創(chuàng)面愈合較糖尿病對(duì)照組及凡士林組愈合速度快,慢于正常對(duì)照組,P0.05;各組創(chuàng)面組織免疫組織化學(xué)染色表明5%阿魏酸組及20mg/kg阿魏酸灌胃組VEGF及TGFβ1含量明顯較糖尿病對(duì)照組及凡士林組高,低于正常對(duì)照組,P0.05;PCR結(jié)果可見(jiàn)5%阿魏酸組及20mg/kg阿魏酸灌胃組VEGF、TGFβ1、HIF-1α表達(dá)較糖尿病對(duì)照組及凡士林組高,低于正常對(duì)照組,P0.05。兔實(shí)驗(yàn)結(jié)果:大體觀察、各組兔耳創(chuàng)面愈合時(shí)間及造創(chuàng)后11天各組創(chuàng)面組織HE染色均表明5%阿魏酸組愈合速度與貝復(fù)濟(jì)組相近、明顯較缺血對(duì)照組及凡士林組快,P0.05;造創(chuàng)后各組創(chuàng)面組織免疫組織化學(xué)染色表明5%阿魏酸組VEGF含量及CD31標(biāo)記血管數(shù)與貝復(fù)濟(jì)組接近,明顯高于缺血對(duì)照組及凡士林組,P0.05;PCR結(jié)果可見(jiàn)5%阿魏酸組VEGF、TGFβ1、HIF-1α表達(dá)與貝復(fù)濟(jì)組接近,明顯高于缺血對(duì)照組及凡士林組,P0.05。結(jié)論:阿魏酸能通過(guò)提高VEGF、TGFβ1及HIF-1α等與新生血管密切相關(guān)的蛋白表達(dá)而促進(jìn)創(chuàng)面血管生成進(jìn)而促進(jìn)慢性創(chuàng)面愈合。
[Abstract]:Objective: to study whether ferulic acid can promote healing of refractory wound and ischemic refractory wound of rabbit ear in diabetic rats. Methods: 20 male Sprague-Dawley rats were set as normal control group. 80 diabetic rats fed with high glucose and high fat diet combined with 45 mg / kg streptozotocin were randomly divided into 4 groups: diabetic control group and vaseline smear group with 5% ferulic acid. Rats in 20mg / kg ferulic acid group were treated with 2 cm 脳 2 cm full-thickness skin defect wounds. The wounds were changed daily and treated with drugs. 40 male New Zealand white rabbits were randomly divided into 4 groups: vaseline group and ischemic control group (5% ferulic acid). The ischemic control group was treated only by dressing. The two animal experiments were carried out at 1 minute after operation (81114). The wound healing rate was calculated at each time point, and the wound healing time was compared. The specimen was divided into two parts from the middle line. The wound healing was observed by hematoxylin-eosin staining and the expression of vascular endothelial growth factor (VEGF) in the tissue was detected by immunohistochemical staining. Results: experimental results: gross observation. The wound healing rate at each time point in each group was higher than that in the control group. The wound healing time and wound tissue HE staining showed that the wound healing rate of 5% mg / kg ferulic acid group and 20 mg / kg ferulic acid group was faster than that of diabetic control group and vaseline group. . It was slower than the normal control group (P 0.05). Immunohistochemical staining of wound tissue showed that the contents of VEGF and TGF 尾 1 in 5% ferulic acid group and 20mg / kg ferulic acid group were significantly higher than those in diabetic control group and vaseline group. It was lower than the normal control group (P 0.05). PCR results showed that the expression of VEGF TGF- 尾 1 HIF-1 偽 in 5% ferulic acid group and 20mg / kg ferulic acid group was higher than that in diabetic control group and vaseline group, and was lower than that in normal control group. P0.05.The results of rabbit experiment showed that the healing time of rabbit ear wound in each group and HE staining of wound tissue in each group 11 days after operation showed that the healing speed of 5% ferulic acid group was similar to that of Befuji group. Compared with ischemic control group and vaseline group, P 0.05 was faster than that in ischemic control group. The immunohistochemical staining of wound tissue in each group showed that the VEGF content and the number of CD31 labeled vessels in the 5% ferulic acid group were similar to those in the Beifuji group, which were significantly higher than those in the ischemic control group and vaseline group (P 0.05). The results of PCR showed that the expression of VEGF TGF- 尾 1 HIF-1 偽 in 5% ferulic acid group was similar to that in epigastric group, and was significantly higher than that in ischemic control group and vaseline group. Conclusion: ferulic acid can promote wound angiogenesis and promote chronic wound healing by increasing the expression of VEGF TGF- 尾 1 and HIF-1 偽.
