表達結核分枝桿菌融合抗原TB10.4-HspX的重組酵母免疫效果研究

【摘要】:結核病是一種嚴重危害人類健康的傳染病,其防治難點主要在于抗藥性菌株的出現(xiàn)以及唯一的抗結核疫苗——卡介苗的效果不理想.為發(fā)展新型抗結核疫苗,本研究中構建了表達結核分枝桿菌融合抗原TB10.4-HspX(TB10H)的重組釀酒酵母,分別以皮下注射和滴鼻兩種方式免疫小鼠,檢測免疫效果.結果顯示,該重組釀酒酵母通過皮下注射的方式免疫后刺激小鼠產生TB10H特異性抗體IgG,分泌Th1型細胞因子IFN-γ,而且重組酵母免疫后在一定程度上刺激小鼠樹突狀細胞的成熟分化和抗原遞呈作用.一系列結果表明,表達融合抗原TB10H的重組釀酒酵母能夠有效激發(fā)小鼠抗原特異性免疫應答.
[Abstract]:Tuberculosis (TB) is a serious infectious disease which is harmful to human health. The prevention and treatment difficulties of TB mainly lie in the emergence of drug-resistant strains and the unsatisfactory effect of the only anti-TB vaccine, BCG (Bacillus Calmette Guerin) vaccine. In order to develop a novel anti-tuberculosis vaccine, recombinant Saccharomyces cerevisiae expressing mycobacterium tuberculosis fusion antigen TB10.4-HspX (TB10H) was constructed in this study. Mice were immunized with subcutaneous injection and nasal drip respectively. The results showed that the recombinant Saccharomyces cerevisiae was immunized by subcutaneous injection to stimulate the production of TB10H specific antibody IgG, to secrete Th1 type cytokine IFN- 緯. Moreover, the recombinant yeast stimulated the maturation, differentiation and antigen presentation of mouse dendritic cells to some extent. A series of results showed that recombinant Saccharomyces cerevisiae expressing fusion antigen TB10H could effectively stimulate the antigen-specific immune response of mice.
