PRU株弓形蟲(chóng)感染小鼠后腦組織免疫病理的研究

【摘要】:目的了解弓形蟲(chóng)PRU株感染小鼠后腦內(nèi)局部細(xì)胞因子對(duì)腦組織的免疫病理作用。方法 51只ICR小鼠分為感染組(33只)和對(duì)照組(18只),感染組經(jīng)腹腔注射弓形蟲(chóng)PRU株10個(gè)包囊/鼠,對(duì)照組腹腔注射同等量的PBS。于感染后第5d、10d、15d、20d、30d和90d,剖殺感染組小鼠5只和對(duì)照組小鼠3只,取腦組織,部分作病理切片;部分提取其RNA,檢測(cè)IFN-γ、IL-4、IL-6和TNF-αmRNA;部分獲取腦組織上清液,用ELISA法測(cè)定上述細(xì)胞因子。結(jié)果相對(duì)于正常對(duì)照組,感染小鼠腦組織中IFN-γ于感染后第5d開(kāi)始升高,第10d時(shí)下降達(dá)到最低,之后開(kāi)始上升;TNF-α在感染第10d或15d時(shí)明顯降低,接著上升并于第30d時(shí)達(dá)到高峰;IL-6在感染第5d時(shí)開(kāi)始升高,感染第10d時(shí)降至最低,之后緩慢上升,并于第30d后達(dá)到高峰;IL-4未見(jiàn)明顯變化。結(jié)論弓形蟲(chóng)PRU感染ICR小鼠的過(guò)程中,腦組織中IFN-γ、IL-6和TNF-α細(xì)胞因子在感染第10d或15d的低表達(dá)有利于弓形蟲(chóng)逃避宿主的免疫殺傷作用,導(dǎo)致速殖子在腦組織中存活;同時(shí)細(xì)胞因子的分泌不平衡,導(dǎo)致小鼠腦組織中出現(xiàn)弓形蟲(chóng)包囊與病理變化共存的隱性感染狀態(tài)。
[Abstract]:Objective to investigate the immunopathologic effects of local cytokines in the brain of mice infected with Toxoplasma PRU strain. Methods 51 ICR mice were divided into infection group (n = 33) and control group (n = 18). The infected group received intraperitoneal injection of Toxoplasma gondii PRU strain 10 cysts per mouse, and the control group received the same amount of PBS. intraperitoneally. At the 5th day, 10 days after infection, 15 days after infection, 20 days after infection, 30 and 90 days after infection, 5 mice in the infected group and 3 mice in the control group were dissected, brain tissues were taken for pathological sections, and IFN- 緯, IL-4,IL-6 and TNF- 偽 mRNA; were partly extracted for detection of IFN- 緯, IL-4,IL-6 and TNF- 偽 mRNA;. The supernatant of brain tissue was obtained and the cytokines were determined by ELISA method. Results compared with the normal control group, the IFN- 緯 in the brain tissue of the infected mice began to increase at the 5th day after infection, reached the lowest level on the 10th day, and then began to rise. TNF- 偽 decreased significantly on the 10th or 15th day of infection, then increased and reached the peak on the 30th day, IL-6 began to increase on the 5th day, reached the lowest level on the 10th day, then increased slowly, and reached the peak at the 30th day after infection. There was no obvious change in IL-4. Conclusion during ICR mice infected with Toxoplasma gondii PRU, the low expression of IFN- 緯, IL-6 and TNF- 偽 cytokines in brain tissue on the 10th or 15th day of Toxoplasma gondii infection is beneficial to the immune killing effect of Toxoplasma gondii on the 10th or 15th day of infection, leading to the survival of tachyzoites in brain tissue. At the same time, the secretion of cytokines was unbalanced, resulting in the occult infection of Toxoplasma gondii cyst and pathological changes in the brain tissue of mice.
