【摘要】：目的對(duì)細(xì)粒棘球蚴病(cystic echinococcosis,CE)患者外周血微小RNA(microRNA,miRNA)進(jìn)行差異性表達(dá)分析,并篩選新型潛在診斷標(biāo)志物。方法提取3例臨床確診的CE患者血清和3名健康者血清miRNA,采用芯片Affymetrix miRNA4.0對(duì)血清中miRNA進(jìn)行高通量篩選,通過生物信息學(xué)分析,找出差異性的miRNA。采用實(shí)時(shí)熒光定量PCR(qRT-PCR)對(duì)21例CE患者血清與25名健康者血清的差異miRNA進(jìn)行驗(yàn)證分析。采用Pearson’s相關(guān)性分析法分析miRNA與包囊重量或子囊個(gè)數(shù)的相關(guān)性。采用IBM SPSS 19.0軟件分析不同miRNA的受試者工作特征曲線(receiver operating characteristic curve,ROC)與曲線下面積(area under curve,AUC)。結(jié)果芯片對(duì)比分析結(jié)果顯示,與健康者血清比較,CE患者血清中有172個(gè)miRNA上調(diào)2倍以上,35個(gè)下調(diào)1/2以下。qRT-PCR結(jié)果表明,hsa-miR-762、hsa-miR-3536、hsa-miR-6125、hsa-miR-6821-5P、hsa-miR-663a、hsa-miR-24-3P、hsa-miR-191-5P、hsa-miR-320a等8個(gè)上調(diào)miRNA的表達(dá)水平分別為10.28±0.38、5.11±0.34、11.02±0.55、11.87±0.58、5.31±0.36、5.75±0.51、5.75±0.51、4.96±0.36,均高于其對(duì)應(yīng)的健康組(P0.05)。ROC曲線分析結(jié)果表明,8個(gè)miRNA的AUC均大于0.8,提示這些分子具有潛在的臨床診斷價(jià)值。相關(guān)性分析結(jié)果顯示,miR-24-3p與包囊重量和子囊數(shù)量均呈正相關(guān)(r=0.525 0,P0.05;r=0.617 5,P0.05)。結(jié)論篩選出的8個(gè)宿主miRNA在CE患者血清中有差異性表達(dá),具有潛在的臨床診斷價(jià)值,其中miR-24-3p還可能與CE的發(fā)生發(fā)展相關(guān)。
[Abstract]:Objective to analyze the differential expression of microRNA (microRNA,miRNA) in peripheral blood of patients with echinococcosis (cystic echinococcosis,CE) and to screen new potential diagnostic markers. Methods the serum miRNA, of 3 patients with clinically diagnosed CE and 3 healthy controls were extracted. The high throughput screening of miRNA in serum was carried out by using Affymetrix miRNA4.0 chip. The difference of miRNA. was found by bioinformatics analysis. Real time fluorescence quantitative PCR (qRT-PCR) was used to verify and analyze the differential miRNA in the sera of 21 patients with CE and 25 healthy controls.
