【摘要】：目的：研究泡球蚴囊液刺激環(huán)境對大鼠肝星狀細胞（HSC-T6）TGF-β1、IL-6、TNF-α表達的影響。 方法：用濃度分別為0、0.01、0.025、0.05、0.1、0.2、0.4、0.9、1.7、3.4、6.8、13.5mg/ml的泡球蚴囊液與大鼠HSC-T6細胞共培養(yǎng)24h，，0mg/ml濃度組做空白對照。ELISA法(酶聯(lián)免疫吸附測定法)檢測各濃度泡球蚴囊液處理組轉(zhuǎn)化生長因子β1（TGF-β1）、白細胞介素6（IL-6）、腫瘤壞死因子-α（TNF-α）的表達變化。 結(jié)果：TGF-β1的表達在泡球蚴囊液濃度為0.01～6.8mg/ml范圍內(nèi)略高于空白對照組，但差異無統(tǒng)計學意義；泡球蚴囊液濃度高于3.4mg/ml時，IL-6的表達極顯著增加（P 0.01），其中6.8、13.5mg/ml泡球蚴囊液處理組相比空白對照組分別增加3028.65、3879.23倍；泡球蚴囊液濃度≤0.2mg/ml時，TNF-α的表達趨于平穩(wěn)；此后急劇增加又逐漸下降，其中泡球蚴囊液濃度為0.4mg/ml時TNF-α的表達量達到最大，為空白對照組的3.53倍，差異具統(tǒng)計學意義（P0.05）。 結(jié)論：泡球蚴囊液對大鼠HSC-T6細胞TGFβ1無明顯的刺激分泌作用；高濃度的泡球蚴囊液可刺激大鼠HSC-T6細胞IL-6的表達分泌；一定濃度的泡球蚴囊液可刺激HSC-T6細胞TNF-α分泌。
[Abstract]:Aim: to study the effect of cyst fluid stimulation on the expression of TNF- 偽 in rat hepatic stellate cells (HSC-T6).
