河南省維生素D受體基因rs2222570和rs731236位點(diǎn)多態(tài)性與結(jié)核病的易感性

【摘要】:目的:探討河南省人群維生素D受體(VDR)基因多態(tài)性和結(jié)核病易感性之間關(guān)系。方法:采用病例對(duì)照研究方法,病例組納入260個(gè)結(jié)核病患者,并以年齡和性別相匹配納入260名健康人作為對(duì)照組。采用PCR-SSCP法對(duì)VDR基因位點(diǎn)多態(tài)性進(jìn)行分析。應(yīng)用SPSS 19.0對(duì)數(shù)據(jù)進(jìn)行統(tǒng)計(jì)分析,SHEsis在線分析2個(gè)位點(diǎn)單倍體型和結(jié)核病發(fā)病的關(guān)聯(lián),MDR軟件3.0.2分析基因和環(huán)境交互作用對(duì)結(jié)核病發(fā)病的影響。結(jié)果:VDR基因rs2222570位點(diǎn)多態(tài)性與結(jié)核病發(fā)病風(fēng)險(xiǎn)相關(guān),野生型C/C降低了結(jié)核病發(fā)病50%的風(fēng)險(xiǎn)(校正OR=0.501,95%CI:0.343~0.709),并且該位點(diǎn)等位基因C在對(duì)照組中高于病例組(校正OR=0.632,95%CI:0.493~0.868);而VDR基因的rs731236位點(diǎn)突變基因型C/C增加結(jié)核病發(fā)病風(fēng)險(xiǎn)(校正OR=2.001,95%CI:1.838~2.191)。VDR基因單倍體型CT、TT和TC與結(jié)核病發(fā)病相關(guān)[CT:OR(95%CI)=0.673(0.521~0.847),P0.001;TT:OR(95%CI)=1.332(1.043~1.711),P=0.032;TC:OR(95%CI)=6.561(2.366~18.143),P0.001]。MDR結(jié)果表明VDR基因rs2222570和rs731236位點(diǎn)多態(tài)性、吸煙、結(jié)核病患者密切接觸史4個(gè)因素交互作用增加了結(jié)核病發(fā)病風(fēng)險(xiǎn)[OR(95%CI)=7.619(2.201~26.354),P0.001]。結(jié)論:VDR基因多態(tài)性與結(jié)核病發(fā)病風(fēng)險(xiǎn)相關(guān),且VDR基因rs2222570和rs731236位點(diǎn)多態(tài)性、吸煙、結(jié)核病患者密切接觸史存在交互作用。
[Abstract]:Objective: to investigate the relationship between vitamin D receptor (VDR) gene polymorphism and susceptibility to tuberculosis in Henan Province. Methods: a case-control study was conducted in 260 patients with tuberculosis and 260 healthy controls matched by age and sex. The polymorphism of VDR locus was analyzed by PCR-SSCP method. The data were statistically analyzed by SPSS 19.0. The correlation between haploid type of two loci and the incidence of tuberculosis was analyzed online by using SPSS 19.0. The effect of interaction between gene and environment on the incidence of tuberculosis was analyzed by using the software 3.0.2. Results the rs2222570 polymorphism of the VDR gene was associated with the risk of tuberculosis. Wild type C / C reduced the risk of TB incidence by 50% (adjusted ORO 0.501 / 95 CI: 0.343 / 0.709), and the allele C of this locus was higher in the control group than in the case group (adjusted ORO 0.63295 CI: 0.493C 0.868), while the rs731236 locus mutation genotype C / C of the VDR gene increased the risk of TB. The haploid type CTTT and TC of the VDR gene were associated with the incidence of tuberculosis. The interaction of four factors, smoking and close contact history of tuberculosis patients, increased the risk of tuberculosis. ConclusionVDR gene polymorphism is associated with the risk of tuberculosis, and the polymorphism of rs2222570 and rs731236 loci of VDR gene, smoking and close contact history of TB patients have interaction.
