人感染的布魯氏菌的檢測(cè)鑒定與MLVA基因分析的研究

【摘要】:目的收集新疆地區(qū)人感染的布魯氏菌,對(duì)其進(jìn)行檢測(cè)、鑒定和MLVA基因分型研究。方法通過(guò)主訴、體征、病原接觸史等收集疑似布魯氏菌病患者62例,對(duì)疑似布魯氏菌病患者進(jìn)行虎紅平板試驗(yàn),進(jìn)行血液培養(yǎng)以分離細(xì)菌,將培養(yǎng)出的細(xì)菌進(jìn)行革蘭染色、觸酶、氧化酶和Vitck2Compact等鑒定試驗(yàn)確定了12株布魯氏菌。使用PCR方法對(duì)分離的12株布魯氏菌進(jìn)行MLVA的16個(gè)位點(diǎn)進(jìn)行檢測(cè),結(jié)果測(cè)序后進(jìn)行拼接、重復(fù)序列分析后進(jìn)行比對(duì)。結(jié)果疑似布魯氏菌病62例有34例表現(xiàn)為虎紅平板試驗(yàn)陽(yáng)性,12株菌通過(guò)微生物鑒定方法確定為馬耳他布魯氏菌。使用MLVA-8分型12株菌全部為羊種,其中有7株為45型,1株42型,一株62型。有三株與已知序列均不相同。使用MLVA-11或MLVA-16分型則沒(méi)有發(fā)現(xiàn)與已知序列完全相同的基因型。結(jié)論MLVA-16方法可以用于布魯氏菌的分型,其中Panel2組具有高分辨率的特性。新疆地區(qū)人感染的布魯氏菌以羊種為主,12株菌的MLVA分型結(jié)果與已登錄序列存在差異,使用MLVA-8分型方法確定三株為新發(fā)現(xiàn)基因型,分別表現(xiàn)為(1-5-3-12-2-3-3-2)和(1-5-3-11-2-3-3-2)兩種基因型。
[Abstract]:Objective to study the detection, identification and MLVA genotyping of Brucella in Xinjiang. Methods Sixty-two suspected brucellosis patients were collected by main complaint, physical signs and history of exposure to pathogens. The patients with suspected brucellosis were tested with tiger red plate, blood was cultured to isolate bacteria, and the bacteria were stained with Gram. Twelve strains of Brucella were identified by thixozyme, oxidase and Vitck2Compact. PCR method was used to detect 16 MLVA loci of 12 Brucella isolates. The results were sequenced, spliced, repeated sequence analyzed and compared. Results 34 of 62 suspected brucellosis cases were identified as Brucella Malta by microbiological identification. Using MLVA-8, 12 strains were all sheep species, of which 7 strains were type 45, 1 strain 42, and one strain type 62. Three of them were different from the known sequences. No genotypes identical to known sequences were found using MLVA-11 or MLVA-16 typing. Conclusion MLVA-16 method can be used for the typing of Brucella, and the Panel2 group has high resolution characteristic. The results of MLVA genotyping of 12 strains of Brucella in Xinjiang were different from those of the registered strains. Using MLVA-8 typing method, three strains of Brucella were identified as newly discovered genotypes, showing two genotypes: 1-5-3-12-2-3-3-2) and 1-5-3-11-2-3-3-2).
